Expression of Tregs and IL-35 in Peripheral Blood of Patients with Newly Diagnosed Acute Myeloid Leukemia

摘要 目的:探讨初诊急性髓系白血病(AML)患者外周血中调节性T细胞(Regulatory T Cells, Tregs)和白介素-35(IL-35)的表达。方法:收集33例初诊AML患者和40例健康体检者的外周血,采用流式细胞术检测受试者外周血中固有免疫和适应性免疫细胞、Tregs的水平,酶联免疫吸附试验(ELISA)方法检测血浆IL-35水平,RT-PCR法检测P35 mRNA和EBI3 mRNA相对表达量,并分析Tregs与IL-35的相关性。结果:初诊AML患者外周血中CD4^(+)T细胞、CD8^(+)T细胞、B细胞以及NK细胞比例较对照组显著降低(P<0.05)。Tregs、IL-35水平以及P35 mRNA、EBI3 mRNA表达量较对照组显著升高(P<0.01)。AML患者外周血中Tregs与IL-35表达水平呈正相关(r=0.668,P<0.01)。结论:AML患者存在免疫功能缺陷,Tregs和IL-35可能参与了AML患者抑制性肿瘤免疫微环境的形成,可能成为AML治疗的新靶点。 Objective: To explore the expression of regulatory T cells(Tregs) and interleukin-35(IL-35) in peripheral blood of patients with newly diagnosed acute myeloid leukemia(AML).Methods: Peripheral blood samples were collected from 33 newly diagnosed AML patients and 40 healthy volunteers.The levels of innate and adaptive immune cells and Tregs were detected by flow cytometry.Plasma IL-35 level was detected by enzyme-linked immunosorbent assay(ELISA).The expression levels of P35 and EBI3 mRNA were detected by RT-PCR.The correlation between the levels of Tregs and IL-35 was analyzed.Results: The proportion of CD4^(+)T cells, CD8^(+)T cells, B cells and NK cells in peripheral blood of newly diagnosed AML patients were significantly lower than those of control group(P<0.05).The levels of Tregs, IL-35, and the expression of P35 mRNA and EBI3 mRNA were significantly higher than that of healthy control group(P<0.01).The Tregs level was positively correlated with plasma IL-35 level(r=0.668,P<0.01).Conclusion: AML patients have a deficiency in immune function, which is characterized by the high levels of Tregs and IL-35 in peripheral blood, and it might be a new target for the treatment of AML.
