益气泻肺汤对慢性心力衰竭大鼠心脏及认知功能的影响

Effects of Yiqi Xiefei Decoction on Heart and Cognitive Function in Rats with Chronic Heart Failure

目的:观察益气泻肺汤对慢性心力衰竭大鼠心脏和认知功能的影响。方法:将24只无特定病原体(SPF)级SD大鼠分为假手术组、模型组和益气泻肺汤组,每组8只。通过冠状动脉结扎术制作心肌梗死后慢性心力衰竭大鼠模型,益气泻肺汤组给予益气泻肺汤13 g/(kg·d)灌胃,假手术组及模型组按100 g体质量1 mL纯净水灌胃,连续给药8周后,采集大鼠舌象,超声检测大鼠左室射血分数(LVEF)、左室短轴缩短率(LVFS),水迷宫实验检测大鼠空间学习及记忆能力,酶联免疫吸附试验(ELISA)检测大鼠血清血管紧张素Ⅱ(AngⅡ)浓度,苏木精-伊红(HE)染色观察大鼠心肌组织镜下形态。结果:与假手术组比较,模型组大鼠心脏LVEF、LVFS水平明显降低,水迷宫逃避潜伏期时间延长,穿越平台次数减少,血清AngⅡ浓度增加,差异均有统计学意义(P<0.05或P<0.01)。与模型组比较,益气泻肺汤组大鼠心脏LVEF、LVFS水平增高,水迷宫逃避潜伏期时间缩短,穿越平台次数增加,血清AngⅡ浓度减少,差异均有统计学意义(P<0.05或P<0.01)。结论:益气泻肺汤能够有效改善大鼠心脏泵血功能,延缓心肌重塑进程,整体改善心脏功能,并提高大鼠认知水平,其机制可能与下调AngⅡ水平有关。

Objective:To observe the effect of Yiqi Xiefei Decoction on the heart and cognitive function of chronic heart failure rats.Methods:Twenty-four SPF SD rats were divided into control group,model group,and Yiqi Xiefei Decoction group,with 8 rats in each group.A rat model of chronic heart failure after myocardial infarction was established by coronary artery ligation.The rats in Yiqi Xiefei Decoction group were gavaged with Yiqi Xiefei Decoction 13 g/(kg·d),and the rats in model group and sham operation group were gavaged with the same amount of normal saline for 8 weeks.The tongue images of rats were collected.Left ventricular ejection fraction(LVEF)and left ventricular fractional shortening(LVFS)were detected by ultrasound.Spatial learning and memory ability of rats were detected by water maze test.Serum angiotensinⅡ(AngⅡ)concentration was detected by enzyme linked immunosorbent assay(ELISA),and microscopic morphology of cardiac tissue was observed by hematoxylin-eosin(HE)staining.Results:Compared with sham operation group,the LVEF and LVFS of model group were significantly decreased,the escape incubation time of water maze was prolonged,the number of crossing the platform was increased,serum AngⅡconcentration was increased,and the difference was statistically significant(P<0.05 or P<0.01).Compared with model group,the LVEF and LVFS of rats in Yiqi Xiefei Decoction group was increased,the escape incubation time of water maze was shortened,the number of crossing the platform was increased,serum AngⅡconcentration was decreased,the differences were statistically significant(P<0.05 or P<0.01).Conclusion:Yiqi Xiefei Decoction can effectively delay the process of myocardial remodeling,improve the overall cardiac function,and enhance the cognitive level of rats.The mechanism may be related to the down-regulation of AngⅡlevel.
