摘要
目的研究大肠埃希菌在模拟失重条件下连续培养14 d的基因表达变化。方法利用Gravite重力控制系统模拟失重条件,并在此条件下连续传代培养大肠埃希菌14 d,通过转录组RNA质检和原核转录组RNA建库后进行转录组测序,检测模拟失重条件下大肠埃希菌差异基因的表达。并通过GO和KEGG分析对差异基因进行功能富集分析。结果模拟失重导致大肠埃希菌基因表达差异,共有229个表达差异显著的基因,差异显著基因的筛选条件为P≤0.05且|log 2(Fold Change)|≥1,其中158个为上调基因,71个为下调基因。GO基因富集分析结果显示,差异基因共富集到3个大类中的14个小类。KEGG分析差异基因表达显示,229个差异基因中有69个基因(上调基因47个,下调基因22个)富集于68条通路,大部分为代谢相关通路,主要以上调基因的富集通路为主。结论模拟失重可引起大肠埃希菌基因表达的变化,从而导致其内部相关代谢通路及生物学性状发生变化。
Objective To study the gene expression changes of Escherichia coli cultured under simulated weightlessness for 14 d.Methods Escherichia coli was subcultured for 14 d under simulated weightlessness by Gravite gravity control system,and transcriptome sequencing was performed after inspection of transcriptome RNA and library construction of prokaryotic transcriptome RNA to detect the expression of differential genes in Escherichia coli under simulated weightlessness.GO and KEGG analysis were used for functional enrichment of differential genes.Results Simulated weightlessness led to the difference of gene expression in Escherichia coli.There were 229 significantly different genes in total,and the screening conditions for significantly different genes were P≤0.05 and|log 2 Fold Change|≥1,158 of which were up-regulated genes and 71 were down-regulated genes.GO gene enrichment analysis showed that the differential genes were enriched into 14 subclasses of the 3 major classes.KEGG analysis of differential gene expression showed that 69 genes(47 up-regulated genes and 22 down-regulated genes)of 229 differential genes were enriched in 68 pathways,most of which were metabolic-related pathways,mainly the enrichment pathways of up-regulated genes.Conclusion Simulated weightlessness can cause changes in gene expression of Escherichia coli,resulting in changes in related metabolic pathways and biological characteristics.
