TCF3在结直肠癌中高表达并促进结直肠癌转移

TCF3 is highly expressed in colorectal cancer and promotes colorectal cancer metastasis

目的探究T细胞因子3(TCF3)在结直肠癌中的表达情况及预后意义,探索TCF3对结直肠癌细胞侵袭、转移能力的影响。方法综合生物信息学分析和实验验证结直肠癌组织中TCF3的mRNA和蛋白质表达水平,评估TCF3预后意义;通过Western blotting方法检测结直肠癌细胞株中TCF3表达情况,并使用慢病毒转染技术构建TCF3过表达、敲减稳转细胞株;Transwell、裸鼠尾静脉注射肺转移实验探索TCF3表达水平对结直肠癌细胞侵袭、转移能力的影响。结果生物信息学分析结果显示TCF3在结直肠癌组织中高表达(P<0.01),并提示不良预后。免疫组化结果提示TCF3在结直肠癌中表达水平显著高于癌旁组织(P<0.05),此外TCF3阳性患者总体生存时间显著短于TCF3阴性组(P<0.01),而TCF3阳性组术后复发率显著高于TCF3阴性组(P<0.01)。Transwell和尾静脉注射肺转移实验证实上调TCF3的表达水平能够促进结直肠癌转移。结论结直肠癌中TCF3显著上调,提示结直肠癌患者预后不良,且TCF3具有促进结直肠癌细胞侵袭、转移作用。

Objective To investigate the expression of T cell factor 3(TCF3)in colorectal cancer(CRC)and its prognostic values,and to explore its effects on the invasion and metastasis ability of CRC cells.Methods The mRNA and protein expression levels of TCF3 in CRC tissues were verified by bioinformatic analysis and experiment to evaluate the prognostic significance of TCF3.Western blotting was conducted to detect the expression of TCF3 in CRC cell lines,and lentivirus transfection was used to construct stably transfected TCF3-overexpressed and TCF3-knockdown CRC cells.The effects of TCF3 expression level on the invasion and metastasis ability of CRC cells were investigated by Transwell assays and lung metastatic models in nude mice by tail vein injection.Results The results of the bioinformatics analysis showed that TCF3 was highly expressed in CRC tissues(P<0.01),which indicated a poor prognosis.Immunohistochemical results revealed that the protein level of TCF3 in CRC tissues was significantly higher than that in paracancer tissues(P<0.05).In addition,the overall survival time of TCF3-positive group was significantly shorter than that of TCF3-negative group(P<0.01),and the postoperative recurrence rate of TCF3-positive group was significantly higher than that of TCF3-negative group(P<0.01).Transwell assays and lung metastatic experiment by tail vein injection confirmed that upregulated TCF3 expression promoted CRC metastasis.Conclusion TCF3 is markedly up-regulated in CRC tissues,suggesting a poor prognosis in CRC patients.TCF3 can promote the invasion and metastasis of CRC cells.
