不同苎麻品种镉胁迫下谷胱甘肽硫转移酶基因的表达特征 被引量:2

Expression Characteristics of Bn-glutathione Transferases Gene in Different Ramie(Boehmeria nivea)Varieties under Cadmium Stress

摘要 为了研究苎麻BnGSTs基因在镉毒害过程中的表达模式,本研究以‘湘苎3号’苎麻转录组文库中的8个BnGSTs基因序列为研究对象,采用RT和Real-time PCR技术,以‘石阡竹根麻’、‘宜春红心麻’、‘富顺青麻’和‘中苎1号’为材料,分析这些BnGSTs基因在镉胁迫条件下的表达特征。结果发现,BnGSTs1、BnGSTs2、BnGSTs4、BnGSTs6、BnGSTs7、BnGSTs8受镉胁迫积极响应,在根中呈显著增强趋势,表明它们可能与苎麻根抗镉胁迫机制有关;BnGSTs5受镉胁迫在根中表达减弱,暗示BnGSTs5通过抑制表达起作用;BnGSTs2和BnGSTs3的表达水平与苎麻体内的镉富集水平呈正相关,暗示BnGSTs2和BnGSTs3能够影响苎麻对镉的富集。8个BnGSTs基因的表达模式呈现基因型差异性,表明BnGSTs基因应答镉胁迫的表达模式存在基因型差异。 In order to study the ramie BnGSTs gene expression patterns in the process of cadmium poisoning,this study’Xiangzhu No.3’transcriptome library eight BnGSTs genes sequence as the research object,uses the RT and real-time PCR technology,with’Shiqianzhugenma’,’Yichunhongxinma’,’Fushunqingma’and’Zhongzhu No.1’as material,analysis the BnGSTs gene expression characteristics of under cadmium stress.The results showed that BnGSTs1,BnGSTs2,BnGSTs4,BnGSTs6,BnGSTs7 and BnGSTs8 responded positively to cadmium stress and showed a significant trend of enhancement in roots,indicating that they might be related to the mechanism of resistance to cadmium stress in ramie roots.The expression of BnGSTs5 decreased in roots under cadmium stress,suggesting that BnGSTs5 plays a role by inhibiting its expression.The expression levels of BnGSTs2 and BnGSTs3were positively correlated with the cadmium enrichment level in ramie,suggesting that BnGSTs2 and BnGSTs3could affect the cadmium enrichment in ramie.The expression patterns of eight BnGSTs genes showed genotypic differences,indicating that the expression patterns of BnGSTs gene in response to cadmium stress had genotypic differences.
