摘要
由原种风信子(Hyacinthus orientalis)培育而来的现代园艺风信子品种资源极其丰富,为揭示风信子园艺品种间的亲缘关系,为新品种选育提供新思路,本研究利用8对引物组合对30个常见园艺风信子品种进行SRAP分子标记测定,使用NTSYSpc2.1软件的UPGMA法构建聚类树状图。30个风信子品种具有较高的遗传多样性,筛选出的8对引物共扩增获得8862条条带,其中多态性条带4692条,多态性比率为52.95%;所有品种的遗传相似系数范围在0.7161~0.9398,平均值为0.8158。以相似系数0.7161为阈值,30个风信子品种可分为两大类群,其中,‘White Pearl’和‘Carnegie’相似系数最大,为0.9390;‘Blue Star’和‘Blue Magic’相似系数最小,为0.7161,聚类结果发现相同色系、倍性的品种几乎聚为一类。研究表明,现代风信子各品种间具有稳定的遗传相似性,同色系、倍性的风信子亲缘关系较近。因此,可选用不同花色和倍性的风信子品种作为亲本进行园艺新品种选育。
The germplasm resources of the protospecies-Hyacinthus orientalis have changed greatly.In order to reveal the genetic relationship between modern hyacinth varieties,providing a new research idea for breeding new horticultural varieties,8 pairs of primers of SRAP molecular markers were used to analyze 30 common horticultural hyacinth varieties,and NTSYSpc2.1 was applied to UPGMA clustering analysis to construct the phylogenetic tree.The 30 hyacinth varieties had high genetic diversity,and 8 pairs of SRAP primers amplified a total of 8862clear bands,of which 4692 were polymorphic,and the percentage of polymorphic bands was 52.95%;The genetic similarity coefficient among 30 hyacinth varieties ranged from 0.7161 to 0.9398,with the average of 0.8158.By fixing the value of 0.7161 as the threshold of similarity coefficient,30 hyacinth varities can be divided into two broad categories.There is the the highest similarity coefficient of 0.9390 between’White Pearl’and’Carnegie’,Whereas between’Blue Star’and’Blue Magic’being the lowest similarity coefficient of 0.7161.The results of cluster analysis showed that the varieties with the same color and ploidy were almost divided into the same category.The research shows that there was stable genetic similarity among modern hyacinth varieties,the hyacinth group has certain relevance with its color or ploidy.Therefore,hyacinth with different color and ploidy can be used as parents in breeding new horticultural varieties.
