Telocytes作为抗胃癌中药潜在作用靶点的探究

目的 探究telocytes (TCs)作为中药抗胃癌作用的潜在靶点。方法 应用透射电子显微镜、苏木精-伊红染色、免疫组化、多重荧光免疫组化技术,对25例人胃部组织中TCs的形态、分布、免疫表型进行分析,对癌组织中TCs的细胞数量进行半定量分析。结果 TCs多分布于胃腺体之间的间质、血管周围、伴行于肌束周边及肌层间质中,胞体多为梭形、扁平状或椭圆形,突起(telopodes, Tps)数目不等,形状略有差异。TCs表达CD34、血小板源性生长因子受体α多肽(platelet-derived growth factor receptor alpha, PDGFR-α)。胃癌组织中TCs的形态及其网状结构发生改变,胃癌组织黏膜下层和肌层中TCs的细胞数量明显低于正常胃组织,差异有统计学意义(P<0.01)。结论 TCs在人胃组织各层均有分布,其免疫表型为CD34^(+)/PDGFR-α^(+)/c-kit-/CD31-,TCs的缺失可能是胃癌发生发展的重要机制,本研究结果为胃癌临床治疗及中药开发潜在靶点的研究提供了实验依据。

Objective: To explore the telocytes(TCs) as a potential target of Traditional Chinese medicine against gastric cancer. Methods: Transmission electron microscopy, HE staining, immunohistochemistry and multiple fluorescence immunohistochemistry were used to analyze the morphology, distribution and immunophenotype of TCs in the gastric tissues of 25 patients, and the number of TCs cells in the carcinoma tissues was semi-quantitatively analyzed. Results: TCs were mostly distributed in the interstitium between gastric glands, around blood vessels, accompanied by the periphery of muscle bundles and in the interstitium of muscularis. The cell bodies were mostly fusiform, flat or elliptical. The number of Tps varies and the shape was slightly different. Immunohistochemistry showed that TCs expressed CD34 and PDGFR-α. The morphology and network structure of TCs in gastric cancer tissues were changed, and the numbers of TCs in the submucosa and muscularis of gastric cancer tissue were significantly lower than those in normal gastric tissue(P<0.01). Conclusion: TCs were distributed in all layers of human gastric tissue, the immunophenotype was CD34^(+)/PDGFR-α^(+)/C-kit-/CD31-. The loss of TCs may be an important mechanism for the occurrence and development of gastric cancer. The results provided experimental basis for the study of potential targets for the clinical treatment of gastric cancer and the TCM development.
