摘要
为探索高粱淀粉的合适提取方法,提高高粱淀粉的纯度,以龙米粮1号高粱为原料,采用碱法提取高粱淀粉,确定了最佳的提取工艺,然后以碱性蛋白酶为酶解剂,对提取的高粱淀粉进行纯化,得到最佳的纯化参数。结果表明,碱法提取最佳工艺条件为NaOH质量分数0.4%,反应温度40℃,反应时间2 h,料液比为1∶20(g∶mL),高粱淀粉的得率为82.54%;酶法纯化的最佳参数为碱性蛋白酶加入量0.6%,料液比1∶10(g∶mL),酶解温度35℃,酶解时间120 min,此时高粱淀粉纯度为99.02%。
In order to find a suitable extraction method of sorghum starch and improve the purity of sorghum starch,this study took Longmiliang No.1 sorghum as raw material,extracted sorghum starch by alkaline method,and determined the best extraction process.Then alkaline protease was used as the enzymatic hydrolysis agent to purify the optimally extracted sorghum starch to obtain the optimal purification parameters.The results showed that the optimal process conditions for alkaline extraction were as follows:NaOH concentration 0.4%,reaction temperature 40℃,reaction time 2 h,solid-liquid ratio 1∶20(g∶mL),and the yield of sorghum starch was 82.54%.The optimal parameters of enzymatic purification were:alkaline protease dosage of 0.6%,solid-liquid ratio of 1∶10(g∶mL),enzymatic hydrolysis temperature of 35℃,and enzymatic hydrolysis time of 120 min.The verification test showed that the purity of sorghum starch was 99.02%.
