柱前衍生法测定岩藻多糖中的岩藻糖含量

Determination of L-fucose in Fucoidan by Pre-column Derivatization

建立了岩藻多糖中岩藻糖的测定方法。采用PMP法和高效液相色谱法检测岩藻多糖中岩藻糖的含量。岩藻多糖经三氟乙酸水解后,用PMP进行衍生化,并采用配有二极管阵列检测器的反相高效液相色谱仪进行测定。结果表明,L-岩藻糖在质量浓度1~500μg/mL与色谱峰面积具有良好的线性关系,相关系数为0.999。用该方法进行了精密度试验、稳定性试验、重复性试验、加标回收率等试验,其RSD分别为0.73%,0.58%,2.68%,5.27%,样品加标的平均回收率为94.7%。该方法测定岩藻糖的含量简便、快捷,结果的稳定性、重复性好,测定结果准确。

Research established the determination method of L-fucose in fucoidan.Method for the determination of L-fucose in fucoidan was using PMP precolumn derivatization and high performance liquid chromatography(HPLC).Fucoidan was hydrolyzed by trifluoroacetic acid derived with PMP.A reversed phase high performance liquid chromatograph equipped with diode array detector was used for determination.The results showed that there was a good linear relationship between the concentration range of 1~500μg/mL and chromatographic peak area,and the correlation coefficient was 0.999.The RSD of precision test,stability test,repeatability test and recovery were 0.73%,0.58%,2.68%,5.27%respectively,and the average recovery was 94.7%.The method was simple,rapid,stable,reproducible and accurate.
