瓯江彩鲤Scarb 1基因敲除自交子一代的体色相关性状观察

Observation on body color related traits in inbred progeny of Scarb 1 knockout mutants in Oujiang color common carp

摘要 为了解瓯江彩鲤基因敲除体在自交繁育过程中的体色遗传和变异情况,对野生型和敲除了Scarb 1基因的敲除型自繁F_(1)全红、粉玉体色瓯江彩鲤个体的鳞片色素细胞进行了观察比较,并测定了其体内的类胡萝卜素含量。结果显示:野生型全红瓯江彩鲤的鳞片含有黄色素细胞、红色素细胞和虹彩细胞,而敲除型全红瓯江彩鲤的鳞片上只有黄色素细胞和虹彩细胞,没有红色素细胞;野生型和敲除型粉玉瓯江彩鲤的鳞片色素细胞均为虹彩细胞,没有黄色素和红色素细胞;野生型全红瓯江彩鲤的肠道、血浆、肌肉和皮肤中的类胡萝卜素含量均显著高于敲除型全红个体(P<0.05);野生型粉玉瓯江彩鲤的肠道、肌肉和鳍条中的类胡萝卜素含量也显著高于敲除型粉玉个体(P<0.05)。研究结果表明,敲除Scarb 1基因影响了瓯江彩鲤体表红色素细胞的生成,该基因在瓯江彩鲤红、白体色调控中发挥作用。 In this study,pigment cells and carotenoid contents of scales from individuals with the WR(whole red)and WW(whole white)coloration of Oujiang color common carp were observed and measured in both the gene edited mutant F_(1) and the wild fish.The results showed that the scales of the wild-type WR Oujiang color common carp contained xanthophore,erythrophore and iridescent cells,whereas the scales of the mutant type WR contained only xanthophore and iridocyste but no erythrophore.Both scale pigmented cells of wild-type and mutant type WW Oujiang color common carp were iridocyste with no xanthophore and erythrophore.Carotenoids in the gut,plasma,muscle,and skin of wild-type WR individuals were all significantly higher(P<0.05)than those of mutant type WR individuals;Carotenoid contented in the gut,muscle,and fin strips of wild-type WW were also significantly higher(P<0.05)than those of mutant type WW.The present study indicated that knock out of Scarb 1 affect the production of red pigment cells on the body surface of Oujiang color common carp,which played a role in the control of red and white coloration in the Oujiang color common carp.
