摘要
目的:建立小酸浆不同药用部位总黄酮的含量测定方法及考察小酸浆总黄酮的最佳提取工艺。方法:以芦丁为对照品,采用分光光度法测定小酸浆不同药用部位中总黄酮的含量;单因素基础上,通过正交试验考察固液比、乙醇体积分数、超声温度及超声时间4个因素对小酸浆叶总黄酮提取率的影响,得到最佳工艺。结果:芦丁分析浓度在10.6~63.6μg/mL范围内与吸光度呈良好的线性关系,回归方程为Y=11.077X-0.0012,R^(2)=0.9998;小酸浆不同药用部位总黄酮含量测定结果高低顺序为:叶(0.87%)>萼(0.37%)>果(0.16%)>茎(0.10%);最佳提取工艺:50%乙醇提取溶剂,固液比为1∶15,80℃超声70 min,此条件下的提取率为2.0%。结论:该方法简单,数据可信,为科学使用小酸浆不同药用部位提供了可靠依据;乙醇超声法可以用于小酸浆叶总黄酮的提取,其中超声温度对总黄酮提取率的影响最大。
Objective:Establish a method for determine the content of total flavonoids in different medicinal parts of Physalis minima and study the optimal extraction process of total flavonoids in Physalis minima.Methods:Using rutin as a control product,spectrophotometric determination of the total flavonoid content in different medicinal parts of Physalis minima.On the basis of single factor,the effects of solid-liquid ratio,ethanol volume fraction,ultrasonic temperature and ultrasonic time on the total flavonoid extraction rate of Physalis minima leaves were investigated by orthogonal experiments.The total flavonoid content of the Physalis minima leaves was determined by ultraviolet-spectrophotometer to obtain the best process.Results:The detected concentration of rutin was well linearly related to the absorbance in the range of 10.6μg/mL~63.6μg/mL,the regression equation is Y=11.077X-0.0012 and R^(2)=0.9998.The total flavonoid content of different medicinal parts of Physalis minima was measured in the order of leaf(0.87%)>calyx(0.37%)>fruit(0.16%)>Stem(0.10%).The optimal extraction process is obtained:50%ethanol as the extraction solvent,thesolid-to-liquid ratio is 1∶15,80℃sonication for 70 minutes,the extraction rate under this condition is 2.0%.Conclusion:The method is simple and the data are credible,which provides a reliable basis for the scientific use of different medicinal parts of Physalis minima;Ethanol sonication can be used for the extraction of total flavonoids from Physalis minima leaves,where ultrasonic temperature has the greatest influence on the extraction rate of total flavonoids.
