摘要
目的:研究小鼠淋巴内皮祖细胞(Lymphoid Endothelial Progenitor Cells,LEPCs)对于淋巴水肿的治疗影响。方法:分离并培养小鼠骨髓LEPC细胞,培养至3~4代,使用流式细胞术以及成管实验对LEPCs进行鉴定。通过手术方法建立小鼠后肢淋巴水肿模型,并测试模型的稳定性。将LEPCs在淋巴水肿部位进行注射,测量小鼠腿围并对淋巴管进行计数,测量LEPCs组和对照组差异。结果:分离的细胞呈纤维样,通过流式细胞术鉴定其为LEPCs,在VEGF-C诱导下可以形成管样结构;通过手术建立淋巴水肿模型,手术肢较健康肢明显增粗,且一个月内稳定存在;将LEPCs在淋巴水肿部位进行注射,发现LEPCs干预组较对照组腿围明显变细且淋巴管计数增高,两组比较差异具有统计学意义(P<0.05)。结论:LEPCs可以缓解淋巴水肿症状。
Objective:The effect of lymphoid endothelial progenitor cells on lymphedema.Methods:Isolated LEPCs were cultured to 3rd generation.LEPCs were authenticated using flow cytometry and tube formation assay.LEPCs were identified by flow cytometry and tube forming experiment.The mouse hindlimb lymphedema model was established by surgical method,and the stability of the model was tested.LEPCs was injected into the site of lymphedema.Leg circumferences and lymphatic vessels were assessed the difference between the control group and LEPCs groups.Results:The isolated cells showed long spindle-shaped fibrocyte-like cell and identified?as LEPCs by flow cytometry.Tubular structures was observed under VEGF-C induction;The mice lower extremity lymphedema model was created by surgery.The operated?limb was thicker compared with the un-operated limb,and it continued for one mouth;LEPCs was administered via lymphedema tissue injection.It was found that the leg circumference was thicker and the number of microlymphatic vessels was higher in the LEPCs group compared with the control group.Conclusion:LEPCs can relieve the symptoms of lymphedema.
