SLC40A1与女性生殖系统恶性肿瘤预后相关性的生物信息学分析

Bioinformatics Analysis of the Correlation between SLC40A1 and Prognosis of Female Reproductive System Malignancies

目的利用生物信息学的方法分析SLC40A1基因在女性生殖系统恶性肿瘤中表达水平以及其表达量与临床预后的相关性。方法SLC40Al基因编码膜铁转运蛋白(ferroportin1,FPN1)。通过GeneCard分析SLC40A1在正常组织分布情况,GEPIA2分析在宫颈癌(cervical cancer,CESC)、卵巢癌(ovarian cancer,OV)、子宫内膜癌(endometrial cancer,UCEC)中SLC40A1变异体的分布。Oncomine、Timer 2.0、GEPIA2分别验证SLC40A1mRNA在CESC、OV、UCEC中的表达,HPA验证SLC40A1蛋白表达的差异。其次采用GEPIA2、HPA研究SLC40A1表达与肿瘤分期、生存率的临床预后相关性。最后将SLC40A1进行蛋白相互作用网络分析,进一步验证其在铁代谢中的作用及不同疾病中的相关性。结果SLC40A1在铁代谢中具有重要作用,在女性生殖系统恶性肿瘤中表达普遍显著下调,SLC40A1低表达与CESC、UCEC临床分期晚、生存率低密切相关。结论SLC40A1在恶性肿瘤组织中较正常组织表达量低,在CESC、UCEC中表达量低预示患者的肿瘤分期越晚,同时生存率越低,是潜在的临床预后标志物。

Objective To analyze the expression level of SLC40A1gene in female reproductive system malignant tumors and its correlation with clinical prognosis using bioinformatics methods.Methods The SLC40Al gene encodes membrane iron transporter protein1(FPN1).The distribution of SLC40A1 in normal tissues was analyzed by GeneCard,and the distribution of SLC40A1 variants in cervical cancer(CESC),ovarian cancer(OV),and endometrial cancer(UCEC)was analyzed by GEPIA2.Oncomine,Timer 2.0 and GEPIA2 were used to verify the expression of SLC40A1mRNA in CESC,OV and UCEC respectively.HPA was used to verify the difference of SLC40A1 protein expression.Secondly,GEPIA2 and HPA were used to study the clinical prognosis correlation of SLC40A1 expression with tumor stage and survival rate.Finally,SLC40A1 was subjected to protein interaction network analysis to further verify its role in iron metabolism and its correlation in different diseases.Results SLC40A1 played an important role in iron metabolism,and its expression was generally significantly down-regulated in female reproductive system malignant tumors.Low expression of SLC40A1 was closely related to late clinical stage and low survival rate of CESC and UCEC.Conclusion The expression of SLC40A1 in malignant tumor tissues is lower than that in normal tissues,and the low expression in CESC and UCEC indicates that the patient′s tumor stage is later and the survival rate is lower,and it is a potential clinical prognostic marker.
