摘要
目的乳腺癌是女性发病率最高的肿瘤之一,容易复发且预后差,目前关于其发病机制仍不十分清楚,迫切需要一种可靠的预后评估策略。方法分别从癌症蛋白质组图谱数据库(the cancer proteome atlas,TCPA)和肿瘤基因组图谱(the cancer genome atlas,TCGA)数据库下载1178例乳腺癌患者对应的蛋白质表达谱和临床信息,采用Kaplan-Meier、Cox回归和Pearson相关分析软件进行分析。结果2个数据库筛选出3个Hub蛋白,并构建了与乳腺癌者总生存期(overall survival,OS)显著相关的蛋白风险特征,同时3个Hub蛋白在癌组织和正常组织中差异表达。功能富集分析表明,3种Hub蛋白可能被认为是乳腺癌治疗的新靶点。结论分析表明,3个Hub蛋白对乳腺癌的发生发展具有驱动作用,可以作为新的治疗方向。
Objective Breast cancer is one of the most common tumors in women,which is prone to relapse and has a poor prognosis.At present,the pathogenesis of breast cancer is still unclear,and a reliable prognostic assessment strategy is urgently needed.Methods The protein expression profiles and clinical information of 1178breast cancer patients were downloaded from the cancer proteome atlas(TCPA)and the cancer genome atlas(TCGA)databases,respectively,and analyzed by Kaplan-Meier,Cox regression and Pearson correlation analysis software.Results Three Hub proteins were screened from the two databases,and the protein risk characteristics significantly correlated with overall survival(OS)of breast cancer patients were constructed.At the same time,the three Hub proteins were differentially expressed in cancer tissues and normal tissues.Functional enrichment analysis showed that three Hub proteins may be considered as new targets for breast cancer treatment.Conclusion The analysis showed that the three Hub proteins had driving effects on the occurrence and development of breast cancer and could be used as new therapeutic directions.
