柴胡三参胶囊对缺血性大鼠心肌细胞钠通道的影响

Effects of Chaihu Sanshen Capsule on sodium channels of cardiomyocytes in ischemic rats

目的观察柴胡三参胶囊对缺血性大鼠心肌细胞钠通道影响,探讨柴胡三参胶囊作用靶点及其防治心律失常作用机制,为缺血性心律失常中医药治疗提供新的实验依据。方法40只SD雄性大鼠随机分为柴胡三参胶囊组30只与空白对照组10只,分别予以等量柴胡三参胶囊及生理盐水灌胃以制备含药血清及空白血清。5%、10%、20%含药及空白血清作用于大鼠心肌细胞,用MTT法筛选柴胡三参胶囊最佳给药浓度。大鼠心肌细胞复苏、换液、传代后分为5组:空白组、模型组、空白血清组、普罗帕酮组、柴胡三参胶囊组。第一组以普通培养基培养,不做特殊处理。后4组分别加入普通培养基、空白血清、普罗帕酮、柴胡三参胶囊含药血清,培育1h后均建立缺血模型,运用Western Blot法检测各组钠通道Nav1.5蛋白表达水平。结果缺血处理后,模型组、空白血清组、普罗帕酮组、柴胡三参胶囊组钠通道Nav1.5蛋白表达均高于空白对照组(P<0.05);与空白血清组对比,普罗帕酮组及柴胡三参胶囊组钠通道Nav1.5蛋白表达均下降(P<0.05);柴胡三参胶囊组钠通道Nav1.5蛋白表达下降程度低于普罗帕酮组(P<0.05)。结论钠通道是柴胡三参胶囊的作用靶点,柴胡三参胶囊能通过钠通道Nav1.5蛋白影响钠离子电流,具有改善心肌缺血症状,防治心律失常发生的作用。

Objective To observe the effect of Chaihu Sanshen Capsule on sodium channel in myocardial cells of ischemic rats,and explore the target of Chaihu Sanshen Capsule and its mechanism of prevention and treatment of arrhythmia,so as to provide new experimental basis for TCM treatment of ischemic arrhythmia.Methods Twenty male SD rats were randomly divided into Chaihu Sanshen Capsule group 30 rats and blank control group 10 rats.The same amount of Chaihu Sanshen Capsule and normal saline were given to prepare the drug containing serum and blank serum respectively;5%,10%,20%drug and blank serum were used to screen the best concentration of Chaihu Sanshen Capsule by MTT method.After resuscitation,fluid exchange and passage,rats were divided into five groups:blank group,model group,blank serum group,propafenone group,Chaihu Sanshen Capsule group.The first group was cultured in ordinary medium without special treatment.In the latter four groups,normal medium,blank serum,propafenone and Chaihu Sanshen Capsule containing serum were added respectively.Ischemia models were established after 1h of incubation.The expression of sodium channel Nav1.5 protein in each group was detected by Western Blot.Results After ischemic treatment,the expression of sodium channel Nav1.5 protein in model group,blank serum group,propafenone group and Chaihu Sanshen Capsule group was higher than that in blank group(P<0.05);compared with blank serum group,the expression of sodium channel Nav1.5 protein in propafenone group and Chaihu Sanshen Capsule group was decreased(P<0.01);the decrease of sodium channel Nav1.5 protein expression in Chaihu Sanshen Capsule group was lower than that in propafenone group(P<0.05).Conclusion Sodium channel is the target of Chaihu Sanshen Capsule.Chaihus sanshen Capsule can affect sodium ion current through sodium channel Nav1.5 protein,which can improve myocardial ischemia symptoms and prevent arrhythmia.
