摘要
[目的]分析番茄果实绿果肩的品种间差异及叶绿素合成相关转录因子对其的调控作用。[方法]选取2份有果肩番茄材料,直观观察果实果肩颜色,利用乙醇法测定果实果肩部位叶绿素含量,采用荧光定量PCR法检测相关转录因子的相对表达量。[结果]在3个不同的发育期中,X115番茄品种的果肩颜色始终较X88深,且叶绿素含量也始终显著高于X88;在5个选择的相关转录因子中,Sl GLK2的表达量最高,并且其变化趋势与叶绿素含量基本一致。[结论 ]番茄果实绿果肩深浅存在品种间差异,且受遗传因素的影响,同时证明了Sl GLK2在其中发挥着重要作用,Sl GLK2可能具有调控叶绿素的合成进而影响果肩颜色深浅的作用。
[Objective] The aim was to analyze the differences of fruit green shoulder among different tomato varieties and the regulatory role of chlorophyll synthesis related transcription factors. [Method] Two tomato materials with fruit shoulder were selected,and the color of fruit shoulder was observed intuitively. The ethanol method was used to determine the chlorophyll content of fruit shoulder. [Result] The fruit shoulder color of X115 was always darker than that of X88 through the three stages of development,and the chlorophyll content of the former was always significantly higher than that of the latter. Among the five selected transcription factors,Sl GLK2 had the highest expression level,and the expression trends were basically consistent with chlorophyll content. [Conclusion] The depth of tomato fruit green shoulder was different among varieties and was affected by genetic factors. It was also proved that Sl GLK2 played an important role in this phenomenon,Sl GLK2 regulated the synthesis of chlorophyll and influenced the depth of fruit shoulder color.
