一种基于等温扩增的便携式核酸检测系统

摘要 针对病原体核酸现场即时检测的需求,研发了一种基于等温扩增的便携式核酸检测系统。主要由温度控制模块、样品移动模块和荧光检测模块组成,可以同时检测16个样品。温度控制模块使用半导体制冷片作为变温元件,控温精度为±0.1℃。样品移动平台基于Core XY平面运动结构,采用同步带传动,显著减轻了检测系统的重量。荧光检测模块采用发光二极管和硅光电二极管作为激发光源和荧光探测器,具有尺寸小、功耗低等特点。上述3个模块均通过下位单片机进行控制,单片机通过串口与具有人机交互功能的上位工控机进行通信,然后利用开发的控制软件进行检测参数的设置。利用研发的系统对黄瓜绿斑驳花叶病毒进行了基于环介导等温扩增的核酸检测实验,在45 min内便获得了检测结果。 A portable nucleic acid detection system based on isothermal amplification was developed for point-of-care testing of nucleic acidsof pathogens.The system mainly consisted of a temperature control module,a sample movement module and a fluorescence detection module,and it could test 16 samples simultaneously.Semiconductor chilling plates were used as the temperature adjusting element of the temperaturecontrol module,and the temperature control accuracy was±0.1℃.The sample movement module took a Core XY planar motion structure,andsynchronous belts were used for transmission,which dramatically decreased the weight of the detection system.The light emitting diode(LED)and silicon photodiode were used as the excitation light source and fluorescence detector of the fluorescence detection module,respectively,and therefore this module had a small dimension and low power consumption.The above three modules were controlled by the lower microcontrol unit(MCU),the MCU communicated with the upper industrial personal computer(IPC)with human-computer interaction functionthrough the serial port,and the test parameters were set by the developed control software.By this system,the cucumber green mottle mosaicvirus was detected based on loop-mediated isothermal amplification(LAMP),and the results were obtained within 45 minutes.
