白花泡桐NCED基因家族分析及其对丛枝植原体的响应 被引量:2

摘要 【目的】研究NCED家族基因在泡桐丛枝病发生过程中的作用。【方法】基于白花泡桐(Paulownia fortune)基因组图谱,采用生物信息学方法对白花泡桐NCED(PfNCED)家族进行成员鉴定,系统进化、基因结构和启动子顺式作用元件进行分析,并结合转录组、实时定量PCR和互作蛋白预测等方法筛选与丛枝病发生相关基因。【结果】发现PfNCED家族共有28个基因,分布在8条染色体上;进化树分析结果表明该家族基因可以分为4组,与拟南芥的同源性较高;共线性分析发现该家族发生5次片段复制事件;顺式作用元件表明PfNCED可能响应植物激素处理和环境胁迫。结合丛枝病发生前后的转录组分析,发现7个差异PfNCED可能与泡桐丛枝病发生相关;进一步的蛋白互作网络分析发现PfNCED16可能在丛枝病腋芽形成中发挥重要作用。【结论】白花泡桐中有28个PfNCED基因,其中PfNCED16可能在白花泡桐丛枝形成过程中发挥重要作用。 【Objective】The purpose of this study is to investigate the role of NCED family genes in the occurrence of Paulownia witches’ broom(PaWB). 【Method】Based on the genome map of Paulownia fortunei, the bioinformatics methods were used to identify the PfNCED gene members and analyze its phylogenetic tree, gene structure and promoter cis-acting elements. At the same times, Transcriptome, real-time quantitative PCR and interaction protein prediction were used to screen the specific PfNCED gene related to PaWB. 【Result】It was found that the PfNCED family had a total of 28 genes that distributed on 8 chromosomes.Phylogenetic tree analysis showed that the genes of this family could be divided into four groups, which had high homology with Arabidopsis thaliana. Collinear analysis showed that 5 fragment duplication events occurred in this family. Cis-acting elements suggested that PfNCED might respond to phytohormone treatments, abiotic and biotic stresses. Combined with the transcriptome analysis before and after the occurrence of PaWB, it was found that 7 genes were significantly differentially expressed, which may be related to the occurrence of PaWB. Further protein interaction network analysis revealed that PfNCED16 may play an important role in axillary bud formation in PaWB. 【Conclusion】There are 28 PfNCED genes in Paulownia, among with PfNCED16 was likely to play an important role in the formation of axillary buds.
