多重生物信息分析预测免疫相关基因FN1为干燥综合征发病核心基因

摘要 目的:利用多重生物信息分析挖掘和预测干燥综合征(SS)发病过程的核心基因及关键通路。方法:GEO数据库下载SS基因数据GSE97614,利用R语言中limma数据包进行均一化数据处理,GEO2R分析筛选差异表达基因(DEGs)。多重生物信息分析软件对GSE97614数据进行分析。DAVID在线软件对DEGs进行GO分析及KEGG信号通路预测。STRING在线软件对DEGs进行蛋白互作(PPI)网络分析。Cytoscape对互作网络结果进行可视化处理,并应用内置软件cytohubba筛选出SS发病的核心基因。结果:GSE97614包含9例SS患者及3例正常对照者,共对32321个基因进行检测,limma数据包均一化数据处理并筛选出96个差异上调和65个差异下调表达的基因。基因热图显示全部DEGs具有可聚类性。GO分析主要富集于细胞外基质、细胞外间隙及整合素结合过程。信号通路主要富集于MAPK通路及VEGF通路。PPI网络显示共有141个基因间存在224条联系,cytohubba计算出FN1、IL1B、TIMP1、THBS1、PLAU、SERPINE1、FOS、ITGB3、VCAN、ADAMTS1为前10位发病核心基因。其中FN1综合分析得分为32分,ADAMTS1综合分析得分为10分。结论:多重生物信息分析发现免疫相关基因FN1为SS发病过程的核心基因,可能作为SS快速诊断和治疗的血清标志物及潜在的药物干预靶点。 Objective:To predict hub genes and key pathways in pathogenesis of Sj?gren’s syndrome(SS)by multiple bioinformatics analysis.Methods:GSE97614 data of SS was downloaded from GEO database.limma data package in R language was used for homogenization data processing and GEO2R was used for screen differentially expressed genes(DEGs).Multiple bioinformatics analysis software was used to analyze GSE97614 data.DAVID online software was used to predict DEGs including GO analysis and KEGG signaling pathway.Protein-protein interaction(PPI)network of DEGs was analyzed by STRING online software.Results:GSE97614 contained 9 patients with SS and 3 normal controls.A total of 32321 genes were detected.There were 96 differentially upregulated and 65 down-regulated genes were screened by limma data package after homogenization data processing.Heat maps showed that all the DEGs could be clustered.GO analysis was mainly concentrated in extracellular matrix,extracellular space and integrin binding process.Signaling pathways were mainly enriched in MAPK pathway and VEGF pathway.PPI network showed that there were224 associations among 141 genes.Cytohubba calculated that FN1,IL1B,TIMP1,THBS1,PLAU,SERPINE1,FOS,ITGB3,VCAN,ADAMTS1 were the top 10 hub genes.Comprehensive analysis score of FN1 was 32 and ADAMTS1 was 10 point.Conclusion:Multiple bioinformatics analysis found that immune related gene FN1 is the core gene during pathogenesis of SS,which may be used as a serum marker for rapid diagnosis and a potential drug intervention target of SS.
