HPLC法同时测定玉女煎中7种成分及化学计量学分析

摘要 目的建立HPLC法同时测定玉女煎中地黄苷D、5-羟甲基糠醛、新芒果苷、芒果苷、β-蜕皮甾酮、毛蕊花糖苷、异毛蕊花糖苷的含量,并进行化学计量学分析。方法该药物的分析采用Welchrom C_(18)色谱柱(250 mm×4.6 mm,5μm);流动相0.1%磷酸-乙腈,梯度洗脱;体积流量1.0 mL/min;柱温30℃;检测波长203、250、258、284、317、334 nm。对7种处理方法所得溶液进行层次聚类分析、主成分分析、正交偏最小二乘判别分析。结果7种成分在各自范围内线性关系良好(r≥0.9997),平均加样回收率96.76%~101.37%,RSD 0.6%~4.1%。不同溶液聚为3类,前3个主成分累积方差贡献率超过90%,芒果苷、新芒果苷、地黄苷D的VIP值大于1。结论该方法简便灵敏,重复性好,可用于玉女煎的质量控制。芒果苷、新芒果苷、地黄苷D可作为主要质量评价指标,在后期制备过程中其含量和转移率应重点关注。 AIM To establish an HPLC method for the simultaneous content determination of rehmannioside D,5-hydroxymethylfurfural,neomangiferin,mangiferin,β-ecdysone,acteoside and isoacteoside in Yunv Decoction and to make chemometric analysis.METHODS The analysis of this drug was performed on a 30℃thermostatic Welchrom C_(18) column(250 mm×4.6 mm,5μm),with the mobile phase comprising of 0.1%phosphoric acid-acetonitrile flowing at 1.0 mL/min in a gradient elution manner,and the detection wavelengths were set at 203,250,258,284,317,334 nm.Hierarchical cluster analysis,principal component analysis and orthogonal partial least square-discriminant analysis were adopted in the solutions obtained by seven processing method.RESULTS Seven constituents showed good linear relationships within their own ranges(r≥0.9997),whose average recoveries were 96.76%-101.37%with the RSDs of 0.6%-4.1%.Different solutions were clustered into three types,the former three principal components demonstrated the accumulative variance contribution rate of above 90%,and the VIP values of mangiferin,neomangiferin and rehmandroside D were more than 1.CONCLUSION This simple,sensitive and reproducible method can be used for the quality control of Yunv Decoction.Mangiferin,neomangiferin and rehmandroside D can be taken as main quality evaluation indices,whose contents and transfer rates should be paid close attention to during post preparation.
