人参皂苷Rd通过调控circDONSON表达对结肠癌SW480细胞增殖和凋亡的影响 被引量:3

摘要 目的探讨人参皂苷Rd是否可通过调控环状RNA DONSON(circDONSON)表达来抑制结肠癌SW480细胞的增殖和凋亡。方法选取30例结肠癌患者癌组织及匹配的癌旁组织,体外培养结肠癌SW480细胞,将其分为对照组,50、100、150μmol/L人参皂苷Rd组,si-NC组,si-circDONSON组,人参皂苷Rd+pcDNA组,人参皂苷Rd+pcDNA-circDONSON组,RT-qPCR法检测circDONSON表达,MTT法检测细胞活力,流式细胞术检测细胞凋亡,Western blot法检测蛋白表达。结果与癌旁组织比较,结肠癌组织中circDONSON表达升高(P<0.05)。与对照组比较,100、150μmol/L人参皂苷Rd组细胞活力、circDONSON表达和CyclinD1、Bcl-2蛋白表达降低(P<0.05),细胞凋亡率和p21、Bax蛋白表达升高(P<0.05),并呈剂量依赖性(P<0.05);与si-NC组比较,si-circDONSON组细胞活力和CyclinD1、Bcl-2蛋白表达降低(P<0.05),细胞凋亡率和p21、Bax蛋白表达升高(P<0.05);与人参皂苷Rd+pcDNA组比较,人参皂苷Rd+pcDNA-circDONSON组细胞活力和CyclinD1、Bcl-2蛋白表达升高(P<0.05),细胞凋亡率和p21、Bax蛋白表达降低(P<0.05)。结论人参皂苷Rd可能通过下调circDONSON表达来抑制结肠癌SW480细胞的增殖和凋亡。 AIM To investigate whether ginsenoside Rd can inhibit the proliferation and apoptosis of colon cancer SW480 cells by regulating the expression of circular RNA DONSON(circDONSON).METHODS Thirty patients colon cancer tissues and matching adjacent healthy tissues were selected.Colon cancer SW480 cells were cultured in vitro and divided into the control group,ginsenoside Rd groups(50,100,150μmol/L),si-NC group,si-circDONSON group,ginsenoside Rd+pcDNA group,ginsenoside Rd+pcDNA-circDONSON group.The cells were subsequently subjected to the determination of circDonson expression by RT-qPCR;assessment of cell viability by MTT method;detection of cell apoptosis by flow cytometry;and determination of protein expression by Western blot.RESULTS Compared with the adjacent healthy tissues,colon cancer tissues was found to be with increased circDONSON expression(P<0.05).Compared with the control group,the 100,150μmol/L ginsenoside Rd groups shared decreases in cell viability,circDONSON expression,CyclinD1 and Bcl-2 protein expressions(P<0.05),and increases in apoptosis rate and expressions of p21 and Bax proteins(P<0.05)in a dose-dependent manner(P<0.05).Compared with the si-NC group,the si-circDONSON group was observed with decreased cell viability,CyclinD1 and Bcl-2 protein expressions,and the apoptosis rate as well as the expressions of p21 and Bax proteins(P<0.05).Compared with the ginsenoside Rd+pcDNA group,the ginsenoside Rd+pcDNA-circDONSON group exhibited increased cell viability,expressions of CyclinD1 and Bcl-2 proteins(P<0.05),and decreased apoptosis rate and expression of p21 and Bax proteins(P<0.05).CONCLUSION Ginsenoside Rd may inhibit the proliferation and apoptosis of colon cancer SW480 cells by down-regulating the circDONSON expression.
