摘要
目的研究盒果藤Operculina turpethum(Linn.)S.Manso化学成分及其抗炎活性。方法盒果藤乙醇提取物采用硅胶、大孔吸附树脂、Sephadex LH-20、MCI、半制备液相色谱进行分离纯化,根据理化性质及波谱数据鉴定所得化合物的结构。采用RAW264.7模型评价其体外抗炎活性。结果从中分离得到12个化合物,分别鉴定为大黄素-6-甲醚(1)、大黄素(2)、芹菜素(3)、山柰酚(4)、木犀草素(5)、车前苷(6)、木犀草素-4′-O-β-D-葡萄糖苷(7)、木犀草素-7-O-β-D-葡萄糖苷(8)、芹菜素-7-O-β-D-葡萄糖苷(9)、木犀草素-4′-O-(6″-O-乙酰基)-β-D-葡萄糖苷(10)、邻苯二甲酸二丁酯(11)、槲皮素(12)。化合物4、6、10质量浓度在12.5μg/mL时对NO的抑制率较高。结论化合物10为新化合物,属于新黄酮酰化糖苷。化合物1~2、4、6、11~12均为首次从该植物中分离得到,化合物4、6、10有较好的抗炎活性。
AIM To study the chemical constituents from Operculina turpethum(Linn.)S.Manso and their anti-inflammatory activities.METHODS The ethanol extract from O.turpethum was isolated and purified by silica gel,macroporous adsorption resin,sephadex LH-20,MCI and semipreparative HPLC,then the structures of obtained compounds were identified by physicochemical properties and spectral data.Their anti-inflammatory activities in vitro were evaluated by RAW264.7 model.RESULTS Twelve compounds were isolated and identified as physcion(1),emodin(2),apigenin(3),kaempferol(4),luteolin(5),plantaginin(6),luteolin-4′-O-β-D-glucopyranoside(7),luteolin-7-O-β-D-glucopyranoside(8),apigenin-7-O-β-D-glucopyranoside(9),luteolin-4′-O-(6″-O-acetyl)-β-glucopyra noside(10),dibutyl phthalate(11),quercetin(12).Compounds 4,6,10 had high inhibitory rates on NO at the concentration of 12.5μg/mL.CONCLUSION Compound 10 is a new compound belonging to flavonoid acylated glycoside.Compounds 1-2,4,6,11-12 are first isolated from this plant.Compounds 4,6,10 show good anti-inflammatory activities.
