摘要
目的研究红毛悬钩子Rubus pinfaensis Levl.et Vant.化学成分及其抗肿瘤活性。方法红毛悬钩子90%乙醇提取物采用硅胶、凝胶和RP-18反相柱色谱进行分离纯化,根据理化性质及波谱数据鉴定所得化合物的结构。采用MTT法测试各化合物对LM3肝癌细胞的抑制作用。结果从中分离得到16个化合物,分别鉴定为β-谷甾醇(1)、β-stostenone(2)、7-ketositosterol(3)、ergosterol peroxide(4)、24-乙基-5α-胆甾-3β-5-6β-三醇(5)、kaempferol-3-β-D-(6-O-trans-p-coumary)glucopyranoside(6)、kaempferol-3-β-D-(6-O-cis-p-coumary)glucopyranoside(7)、blumenol A(8)、号角树酸3-甲酯(9)、齐墩果酸(10)、红大戟酸A(11)、阿江榄仁树葡糖苷Ⅱ(12)、2α,3β,19α-三羟基齐墩果烷-12-烯-28-O-β-D-吡喃葡萄糖苷(13)、rubuside B(14)、2α,3β-dihydroxyurs-12,18-dien-28-oic acid 28-O-β-D-glucopyranosyl ester(15)、乌苏酸(16)。化合物14~16 IC 50分别为(28.13±1.45)、(27.36±1.44)、(16.2±1.32)μmol/L。结论化合物1~9、11~15均为该植物中首次分离得到,化合物14~16具有较好的抗肿瘤活性。
AIM To study the chemical constituents from Rubus pinfaensis Levl.et Vant.and its anti-tumor activity.METHODS The 90%ethanol extract from R.pinfaensis was isolated and purified by silica gel,sephadex LH-20 and RP-18 column chromatography,then the structures of obtained compounds were identified by physicochemical properties and spectral data.MTT assay was used to test the inhibitory effects of various compounds on LM3 liver cancer cells.RESULTS Sixteen compounds were isolated and identified asβ-stosterol(1),β-stostenone(2),7-ketositosterol(3),ergosterol peroxide(4),24-ethyl-5α-cholestane-3β-5-6β-triol(5),kaempferol-3-β-D-(6-O-trans-p-coumary)glucopyranoside(6),kaempferol-3-β-D-(6-O-cis-p-coumary)glucopyranoside(7),blumenol A(8),cecropiacic acid 3-methyl ester(9),oleanolic acid(10),knoxivalic acid A(11),arjunglucosideⅡ(12),2α,3β,19α-trihyhydroxy-olean-12-en-28-O-β-D-glucopyranoside(13),rubuside B(14),2α,3β-dihydroxyurs-12,18-dien-28-oic acid 28-O-β-D-glucopyranosyl ester(15),ursolic acid(16).Compounds 14-16 had obvious inhibitory effects with the IC 50 values of(28.13±1.45),(27.36±1.44),(16.2±1.32)μmol/L.CONCLUSION Compounds 1-9,11-15 are first isolated from this plant.Compounds 14-16 show good anti-tumor activities.
