摘要
目的:研究光质对刺五加根中初生代谢产物及初生代谢途径的影响,进而揭示光质对刺五加根的初生代谢调控机制,为刺五加的人工培育提供理论依据。方法:利用气相色谱-质谱联用(GC-MS)技术对自然光、红光、黄光和蓝光4种光质处理下刺五加根初生代谢特征进行系统分析。结果:利用GC-MS技术在不同光质处理的刺五加根中鉴定出的39种初生代谢产物,主要为有机酸、糖类及醇类物质;通过多元统计分析,共筛选到不同光质处理间的28种差异代谢物,差异代谢物主要富集于丙氨酸、天冬氨酸和谷氨酸代谢、半乳糖代谢、柠檬酸循环代谢途径。红光可显著促进丙氨酸、天冬氨酸和谷氨酸代谢和柠檬酸循环代谢途径中代谢物的累积,蓝光可显著促进半乳糖代谢中醇类代谢物的累积。结论:不同光质处理可调控刺五加根的初生代谢途径,影响初生代谢产物的累积。
Objective:To study the effects of light quality on the primary metabolites and primary metabolic pathways in the roots of Acanthopanax senticosus,and then to reveal the regulation mechanism of light quality on the primary metabolism,so as to provide a theoretical basis for the artificial cultivation of the plant.Methods:The primary metabolic characteristics of Acanthopanax senticosus root intervened by natural light,red light,yellow light and blue light were systematically analyzed by gas chromatography-mass spectrometry(GC-MS).Results:39 primary metabolites were identified by GC-MS in the roots of Acanthopanax senticosus intervened by different light quality,which were mainly organic acids,saccharides and alcohols.Through multivariate statistical analysis,28 differential metabolites were screened after different light quality interventions,which were mainly concentrated in the pathways of the mechanisms of alanine,aspartic acid and glutamate,as well as cycle metabolisms of galactose and citric acid.Red light significantly promoted the accumulation of metabolites in the metabolic pathways of alanine,aspartic acid,glutamate and citric acid cycle,whereas blue light significantly promoted the accumulation of alcohols in galactose metabolism.Conclusion:Different light qualities regulate the primary metabolic pathways of Acanthopanax senticosus root and affect the accumulation of primary metabolites.
