基于多指标含量测定结合化学计量学评价的石见穿质量研究 被引量:3

摘要 目的 以酚酸类成分及水溶性浸出物含量为指标,结合聚类热图、灰色关联度分析与熵权TOPSIS分析等化学计量学方法综合评价不同产地石见穿药材的质量。方法 参照2020年版《中国药典》中的规定测定水溶性浸出物。建立UPLC法测定石见穿药材中丹酚酸B、丹参素、咖啡酸、绿原酸及迷迭香酸5种酚酸类成分含量,采用Waters BEH C_(18)色谱柱(2.1 mm×100 mm,1.7μm);以0.1%甲酸(A)—乙腈(B)为流动相进行梯度洗脱;检测波长为290 nm;柱温为40℃;流速为0.4 mL/min;进样量为1μL。测定来自广东、广西、山东、安徽、湖北5个产地共20批药材的水溶性浸出物及5种酚酸类成分含量。采用聚类热图、灰色关联度分析与熵权TOPSIS分析对测定结果进行综合评价。结果 不同批次石见穿药材各指标成分含量存在较大差异;聚类热图分析显示20批药材可分成5类;灰色关联度分析与熵权TOPSIS分析均显示产自山东产区的药材其得分排名整体靠前。结论 本研究为石见穿药材的质量评价及产地选择提供参考。 Objective To comprehensively evaluate the quality of Salvia chinensis herbs of different origins using phenolic acid components and water-soluble extracts content as indicators,combined with chemometric methods such as cluster heat map,gray correlation analysis and entropy TOPSIS.Methods The water-soluble extracts was determined with reference to the provisions in the Chinese Pharmacopoeia 2020.UPLC method was established to determine the contents of five phenolic acids,namely,salvianolic acid B,salvianic acid A,caffeic acid chlorogenic acid and rosmarinic acid.Using Waters BEH C_(18)column (2.1 mm×100 mm,1.7μm),gradient elution was performed with 0.1% formic acid (A)-acetonitrile (B) selution as mobile phase;the flow rate was 0.40 mL/min;the detection wavelength was 290 nm;the column temperature was 40℃;the injection volume was 1μL.The content of water-soluble extracts and five phenolic acid components of 20 batches of herbs from five origins in Guangdong,Guangxi,Shandong,Anhui and Hubei were determined.Cluster heat map,gray correlation analysis and entropy TOPSIS were used to comprehensively evaluate the determination results.Results The results showed that there were significant differences in the contents of each index component among different batches of herbs.Cluster heat map analysis showed that 20 batches of medicinal materials could be divided into 5 categories The results of grey correlation analysis and entropy TOPSIS showed that the scores of the medicinal materials from Shandong were the highest overall.Conclusion The study provides reference for the quality evaluation and origin selection of Salvia chinensis.
