小胸鳖甲Attacin基因结构与功能生物信息学分析 被引量:1

摘要 为了解小胸鳖甲抗菌肽基因MpAtt1和MpAtt2的结构与功能,借助生物信息学相关软件,对2个基因所编码的氨基酸理化性质、信号肽、跨膜结构、蛋白质的二级结构,蛋白质翻译后修饰、功能结构域、蛋白质的三级结构及亚细胞定位、Attacin同源性进行生物信息学分析。MpAtt1和MpAtt2二级结构以无规则卷曲和β-折叠为主,存在大量的潜在磷酸化位点,三级结构具有高度保守性,具有Attacin_C结构域和Gloverin结构域,属于Attacin_C超家族。MpAtt1和MpAtt2蛋白为可溶性分泌蛋白,含有N-端信号肽。与鞘翅目光滑鳖甲抗菌肽Attacin1序列在氨基酸水平上具有84.77%的一致性。这表明MpAtt1和MpAtt2具有较好的抗菌活性,为该基因的功能鉴定提供理论基础,以期为研究昆虫小胸鳖甲的耐寒机制提供基因来源。 This study aimed to understand the structure and function of the antimicrobial peptide genes MpAtt1 and MpAtt2 of Microdera punctipennis.Physicochemical properties of amino acids,the signal peptide,transmembrane structure,protein secondary structure,protein posttranslational modification,functional structural domain,protein tertiary structure and subcellular localization,homology analysis of amino acid encoded by the two Attacin genes were conducted bioinformatics analysis with the help of some bioinformatics software.The secondary structures of MpAtt1 and MpAtt2 were dominated by random coil andβ-sheet,with a large number of potential phosphorylation sites,and the tertiary structures were highly conservative with Attacin_C domain and Gloverin domain,belonging to the Attacin_C superfamily.MpAtt1 and MpAtt2 proteins were soluble secretory proteins,containing N-terminal signal peptide.The two Attacin genes showed 84.77%identity with Attacin1 gene of Anatolica polita at the amino acid level.It was shown that MpAtt1 and MpAtt2 had good antibacterial activity,providing a theoretical basis for the functional identification of this gene,with a view to providing a gene source for the study of the cold tolerance mechanism of Microdera punctipennis.
