针刺脾经腧穴对类风湿关节炎模型大鼠关节滑膜细胞超微结构及IL-1β、IL-4、IL-10的影响 被引量:3

Effects of acupuncture at acupoints of spleen meridian on ultrastructure of joint synovial cells,IL-1β,IL-4 and IL-10 in rheumatoid arthritis model rats

摘要 目的观察针刺脾经腧穴阴陵泉及三阴交对类风湿关节炎(RA)模型大鼠关节滑膜细胞超微结构及白细胞介素(IL)-1β、IL-4、IL-10的影响。方法从48只雄性Wistar大鼠中随机抽取12只作为空白组,其余36只作为造模组。采用风、寒、湿环境因素干预并结合完全弗氏佐剂足跖部注射建立RA大鼠模型,造模成功后将36只造模大鼠采用随机数字表法分为模型组、雷公藤组和针刺组,每组12只。针刺组采用掀针针刺阴陵泉与三阴交穴,连续针刺6 d,休息1 d;雷公藤组以8 mg/kg剂量灌胃雷公藤多苷片混悬液,每日1次。2组均以7 d为1个疗程,共治疗3个疗程。空白组与模型组不予干预。比较各组大鼠足趾关节滑膜细胞内细胞器的形态学变化及血清IL-1β、IL-4及IL-10含量。结果与空白组比较,模型组大鼠细胞核核膜间隙增宽,细胞膜破裂,可见细胞器外溢,线粒体空泡变性,粗面内质网明显扩张、断裂,核糖体大量减少;针刺组及雷公藤组上述病理变化均得到一定程度的改善。与空白组比较,模型组大鼠血清IL-1β含量明显升高,IL-4及IL-10含量明显降低(P<0.01);与模型组比较,雷公藤组及针刺组大鼠血清IL-1β含量明显降低,IL-4及IL-10含量明显升高(P<0.01)。结论针刺脾经腧穴不仅可以改善RA模型大鼠局部滑膜细胞的超微结构,还可影响血清中炎症因子的表达。 Objective To observe the effect of acupuncture at acupoints of Yinlingquan(SP9)and Sanyinjiao(SP6)of spleen meridian on ultrastructure of joint synovial cells,interleukin(IL)-1β,IL-4 and IL-10 in rheumatoid arthritis(RA)model rats.Methods Twelve Wistar rats were selected randomly from 48 male ones as blank group,and the remaining 36 ones were used as modeling group.The RA rat model was established by intervening wind,cold and wet environmental factors combined with complete Freund’s adjuvant injection into the soles of the feet.After successful modeling,36 model rats were divided randomly into model group,tripterygium wilfordii group,and acupuncture group,with 12 rats in each group.The acupuncture group received acupuncture at Yinlingquan(SP9)and Sanyinjiao(SP6)by lifting acupuncture for 6 days and rested for 1 day.The tripterygium wilfordii group was given 8 mg/kg tripterygium glycosides tablet suspension by gavage,once a day.Both groups took 7 days as a course of treatment,with a total of 3 courses of treatment;the blank group and the model group were not intervened.The morphological changes of intracellular organelles and the levels of serum IL-1β,IL-4 and IL-10 in the synovial cells of the toe joints of the rats in each group were compared.Results Compared with the blank group,the gap between the nuclear and nuclear membranes of the rats in the model group was widened,partially disappeared,the cell membrane was ruptured,the extracellular organelles were seen,mitochondrial vacuoles were degenerated,the rough endoplasmic reticulum was expanded and broken obviously,and the ribosomes was reduced greatly;while the above pathological changes of the rats in the acupuncture group and tripterygium wilfordii group were improved to some extent.Compared with the blank group,the serum IL-1βcontent of the rats in the model group was increased significantly,while the IL-4 and IL-10 contents were decreased significantly(P<0.01).Compared with the model group,The content of IL-1βin the rats of the tripterygium wilfordii group and acupuncture group were decreased significantly,and the content of IL-4 and IL-10 were increased significantly(P<0.01).Conclusion Acupuncture at acupoints of spleen meridian can not only improve the ultrastructure of local synovial cells in RA model rats,but also affect the expression of inflammatory factors in serum,which provides experimental evidence for the anti-inflammatory and immune effects of acupuncture at acupoints of spleen meridian.
