摘要
目的 研究芦荟皮层外泌体样囊泡(rAEVs)在创面修复中的作用。方法 通过激光共聚焦成像和流式细胞分析验证rAEVs被细胞摄取的可行性。通过自由基清除及细胞内活性氧水平测定实验,验证rAEVs的抗氧化作用。采用划痕和Transwell实验验证rAEVs对细胞迁移的影响。构建小鼠全层皮肤切除模型,将rAEVs作用于创面,验证其对创面的修复作用。结果 rAEVs可经由巨胞饮和膜融合等多种途径被人永生化表皮细胞摄取,具有良好的阳离子自由基及活性氧清除作用,可有效促进细胞增殖和迁移,将其应用于小鼠创面模型治疗14 d后,创面修复率达75.29%。结论 rAEVs可促进创面愈合疗效,有望成为一种经济、有效、安全的新型绿色药物。
Objective To investigate the role of aloe rind derived exosomal-like vesicles(rAEVs) in wound repair. Methods The feasibility of rAEVs uptake was verified by confocal laser imaging and flow cytometry analysis. The antioxidant effect of rAEVs was verified by free radical scavenging and intracellular reactive oxygen species level measurement. Scratch and Transwell experiments were performed to verify the effect of rAEVs on cell migration. Finally, a mouse full-thickness skin excision model was constructed, and rAEVs was applied to the wound to verify whether it had a healing effect on the wound. Results rAEVs could be taken up by HaCaT cells through a variety of ways, such as macrocytopenia and membrane fusion, and had good cationic free radical and reactive oxygen species scavenging effects, which could effectively promote cell proliferation and migration. After 14 days of treatment, the wound healing rate of rAEVs in mouse wound model reached 75.29%. Conclusion rAEVs has better effect on wound healing, and is expected to be an economical, effective and safe new green drug for wound healing treatment.
