狗头赤芍质量标准的提升研究

Research on the Improvement of Quality Standard of Radix Paeoniae Mairei

目的:高效液相色谱(HPLC)法同时测定狗头赤芍中没食子酸和芍药苷的含量,为狗头赤芍质量标准的提升提供依据。方法:采用十八烷基硅烷键合硅胶为填充剂的色谱柱,流动相为乙腈-0.1%磷酸溶液,梯度洗脱,流速为1 mL/min,检测波长为230 nm,柱温为30℃,进样量为10μL。结果:线性和范围分别为,没食子酸:Y=22472X+2118.9,r=0.9999(n=5),在4.237~33.884μg范围内线性关系良好;芍药苷:Y=13583X-12485,r=0.9999(n=5)在5.197~41.578μg范围内线性关系良好。没食子酸的平均回收率为99.58%,RSD为1.17%;芍药苷的平均回收率为99.84%,RSD为0.45%。结论:建立的方法简便快捷、重复性良好、准确度较高、分离度较好,可用于狗头赤芍中没食子酸和芍药苷的含量测定。

Objective Simultaneous determination of gallic acid and paeoniflorin in Radix Paeoniae Aairei by high performance liquid chromatography(HPLC),Provide a basis for the improvement of the quality standard of Radix Paeoniae Aairei.Methods Columns using octadecylsilane-bonded silica as filler,The mobile phase is acetonitrile-0.1%phosphoric acid solution,gradient elution,The flow rate is 1mL/min,Detection wavelength is 230nm,The column temperature is 30℃,The injection volume is 10μL.Results Linear and range are,Gallic acid:Y=22472X+2118.9,r=0.9999(n=5),The linear relationship is good in the range of 4.237~33.884μg;Paeoniflorin:Y=13583X-12485,r=0.9999(n=5),The linear relationship is good in the range of 5.197~41.578μg.The average recovery rate of gallic acid was 99.58%,and the RSD was 1.17%;The average recovery of paeoniflorin was 99.84%,and the RSD was 0.45%.Conclusion The established method is fast,simple,with good repeatability,high accuracy and good separation.It can be used for the quality control of gallic acid and paeoniflorin in PRadix Paeoniae Aairei.
