HPLC法测定不同商品规格桂林产钩藤中4种生物碱的含量 被引量:3

Content Determination of Four Alkaloids in Different Commercial Specifications of Uncariae Ramulus cum Uncis from Guilin by HPLC

摘要 [目的]测定不同商品规格桂林产钩藤中钩藤碱、异钩藤碱、去氢钩藤碱和异去氢钩藤碱的含量。[方法]采用岛津Inertsil ODS3色谱柱(4.6 mm×250 mm,5μm),流动相为0.1%三乙胺(A)-乙腈(B),流速为1.0 ml/min,检测波长为245 nm,进样量为10μl;测定双钩、单钩、混钩(一级、二级)、钩藤枝等不同商品规格桂林产钩藤中钩藤碱、异钩藤碱、去氢钩藤碱和异去氢钩藤碱4种生物碱的含量。[结果]钩藤碱含量由高到低依次是:钩藤枝>单钩>混钩(二级)>双钩>混钩(一级);异钩藤碱含量由高到低依次是:钩藤枝>单钩>混钩(二级)>双钩>混钩(一级);去氢钩藤碱含量由高到低依次是:双钩>混钩(一级)>混钩(二级)>钩藤枝>单钩;异去氢钩藤碱含量由高到低依次是:双钩>混钩(一级)>混钩(二级)>单钩>钩藤枝;四种生物碱总含量由高到低依次是:双钩>混钩(一级)>钩藤枝>混钩(二级)>单钩。[结论]本研究所用方法简便快捷,可为市场划分钩藤不同商品规格提供参考依据。 [Objective]To determine the contents of rhynchophylline,isorhynchophylline,corynoxeine and isoco⁃rynoxeine in Uncariae Ramulus cum Uncis produced in Guilin with different commercial specifications.[Methods]A Shimadzu Inertsil ODS3 column(4.6 mm×250 mm,5μm)was used with a mobile phase of 0.1%triethylamine(A)-ace⁃tonitrile(B),a flow rate of 1.0 ml/min,a detection wavelength of 245 nm and a sample volume of 10μl.The contents of four alkaloids in different commercial specifications of Uncariae Ramulus cum Uncis,such as double hook,single hook,mixed hook(primary and secondary),and branches,including rhynchophylline,isorhynchophylline,corynoxeine and isocorynoxeine were determined.[Results]The content of rhynchophylline in descending order,from high to low,listed as follows:branch>single hook>mixed hook(secondary)>double hook>mixed hook(primary);isorhynchophyl⁃line content:branch>single hook>mixed hook(secondary)>double hook>mixed hook(primary);corynoxeine content:double hook>mixed hook(primary)>mixed hook(secondary)>branch>single hook;isocorynoxeine content:double hook>mixed hook(primary)>mixed hook(secondary)>single hook>branch;total content of the four alka⁃loids:double hook>mixed hook(primary)>branch>mixed hook(secondary)>single hook.[Conclusion]The method applied in this study is simple and quick,which can provide a reference for the market to divide the commercial specifi⁃cations of different Uncariae Ramulus cum Uncis.
