基于指纹图谱及多元统计分析评价鲜人参的质量

Quality Evaluation of Fresh Ginseng Based on Fingerprint Combined with Multivariate Statistical Analysis

摘要 目的:建立鲜人参药材HPLC指纹图谱方法,结合多元统计分析及多组分含量测定评价鲜人参样品的质量。方法:采用Thermo Hypersil BDS C_(18)色谱柱(250 mm×4.6 mm, 5μm);流动相为乙腈(A)-水(B),梯度洗脱;流速:1.0 ml·min^(-1),检测波长:203 nm,进样量:10μl;对照品鉴定7个共有峰,并对4个差异标志物进行定量分析;采用SIMCA-P 14.1软件对34批鲜人参样品进行聚类分析(CA)、主成分分析(PCA)和偏最小二乘法判别分析(PLS-DA)。结果:34批鲜人参样品与对照图谱的相似度均≥0.946,标定了12个共有峰,鉴定7个色谱峰;HCA和PCA将34批鲜人参样品分为2类,筛选出人参皂苷Re、人参皂苷Rf、人参皂苷Rb1和人参皂苷Rd 4个影响分组差异较大的组分。结论:该研究进一步弥补了鲜人参质量评价标准,可为鲜人参的药效物质基础研究提供参考。 Objective: To establish an HPLC fingerprint method for fresh ginseng and evaluate the quality of fresh ginseng samples by multivariate statistical analysis and multicomponent content determination. Methods: A Thermo Hypersil BDS Ccolumn(250 mm×4.6 mm, 5 μm) was used. The mobile phase consisted of acetonitrile(A) and water(B) with gradient elution. The flow rate was 1.0 ml·min^(-1), the detection wavelength was 203 nm, and the injection volume was 10 μl. Seven common peaks were identified and four differential markers were quantitatively analyzed. Cluster analysis(CA), principal component analysis(PCA) and partial least squares discriminant analysis(PLS-DA) were performed on 34 batches of fresh ginseng samples using SIMCA-P 14.1 software. Results: The similarity between 34 batches of fresh ginseng samples and the control spectrum was ≥0.946, 12 common peaks were calibrated and 7 chromatographic peaks were identified;the 34 batches of fresh ginseng samples were divided into 2 groups by HCA and PCA, and ginsenoside Re, ginsenoside Rf, ginsenoside Rband ginsenoside Rd were screened as differential markers. Conclusion: The study further improves the quality evaluation standard for fresh ginseng, and can provide reference for the basic research on pharmacodynamic substances of fresh ginseng.
