摘要
目的:探讨滋膵通脉饮对糖尿病心肌病大鼠糖脂代谢及氧化应激的影响。方法:从51只SD大鼠中随机抽取9只为空白对照组,其余大鼠复制2型糖尿病模型,将造模成功的大鼠随机分为模型组、Rx抑制剂组、滋膵通脉饮高剂量组、滋膵通脉饮低剂量组,每组9只。各给药组大鼠予以相应药物灌胃,空白对照组和模型组均灌服等体积蒸馏水,2次/d,共4周。检测各组大鼠体质量、空腹血糖(FPG)、胰岛素(FINS)、甘油三酯(TG)、总胆固醇(T-CHO)、低密度脂蛋白(LDL-C)、胰岛素抵抗指数(HOMA-IR)、糖化血红蛋白(HbAlc)、超氧化物歧化酶(SOD)及丙二醛酸(MDA),同时应用蛋白免疫印迹法检测大鼠心肌组织蛋白激酶C-βⅡ(PKC-βⅡ)表达情况,苏木精-伊红(HE)染色法观察心肌组织的病理变化。结果:与空白对照组比较,模型组大鼠体质量及SOD活性均明显降低(P<0.05),FPG、FINS、HOMA-IR、TG、T-CHO、LDL-C、HbAlc及MDA指标均明显升高(P<0.05)。与模型组比较,滋膵通脉饮高、低剂量组大鼠体质量、SOD活性均升高(P<0.05),FPG、FINS、HOMA-IR、TG、T-CHO、LDL-C、HbAlc及MDA指标均明显降低(P<0.05),Rx抑制剂组大鼠SOD活性升高(P<0.05),MDA含量明显降低(P<0.05)。与Rx抑制剂组比较,滋膵通脉饮高、低剂量组大鼠体质量明显升高(P<0.05),FPG、FINS、HOMA-IR、HbAlc、TG、T-CHO及LDL-C指标均明显降低(P<0.05);滋膵通脉饮高、低剂量组大鼠SOD活性及MDA含量与Rx抑制剂组比较,差异无统计学意义(P>0.05)。模型组大鼠心肌纤维与心肌细胞排列紊乱,纤维间隙明显增宽,心肌细胞肥大变形、可见炎症细胞浸润、间质纤维化明显。与模型组比较,滋膵通脉饮高、低剂量组及Rx抑制剂组心肌细胞排列、心肌细胞形态及心肌纤维间隙改善。与空白对照组比较,模型组大鼠心肌组织PKC-βⅡ蛋白相对表达量明显升高(P<0.01);与模型组比较,滋膵通脉饮低剂量组及Rx抑制剂组大鼠心肌组织PKC-βⅡ蛋白相对表达量明显降低(P<0.01);滋膵通脉饮高剂量组大鼠心肌组织PKC-βⅡ蛋白相对表达量与模型组比较,差异无统计学意义(P>0.05)。结论:滋膵通脉饮能改善糖尿病心肌病大鼠糖脂代谢及氧化应激,同时可抑制PKC-βII表达抗心肌纤维化。
Objective:To investigate the effect of Zicui Tongmai Yin on glucose and lipid metabolism and oxidative stress in rats with diabetes cardiomyopathy.Methods:9 SD rats were randomly selected from 51 SD rats as the blank control group,and the rest of the rats were duplicated into type II diabetes models.The rats that were successfully modeled were randomly divided into model group,Rx inhibitor group,Zicui Tongmai Yin high-dose and low-dose group,with 9 rats in each group.Rats in each treatment group were given corresponding drugs by gavage,and the blank control group and model group were given equal volume of distilled water by gavage twice a day for 4 weeks.The body mass,fasting blood glucose(FPG),insulin(FINS),triglyceride(TG),total cholesterol(T-CHO),low density lipoprotein(LDL-C),insulin resistance index(HOMA-IR),glycosylated hemoglobin(HbAlc),superoxide dismutase(SOD)and malonic acid(MDA)of rats in each group were measured,and the expression of protein kinase C-βII(PKC-βII)in myocardial tissue of rats was detected by Western blotting,the pathological changes of myocardial tissue were observed by hematoxylin-eosin(HE)staining.Results:Compared with the blank control group,the body mass and SOD activity of rats in the model group were significantly decreased(P<0.05),and the indexes of FPG,FINS,HOMA-IR,TG,T-CHO,LDL-C,HbAlc and MDA were significantly increased(P<0.05).Compared with the model group,the body mass and SOD activity of rats in Zicui Tongmai Yin high and low dose groups were increased(P<0.05),and the indexes of FPG,FINS,HOMA-IR,TG,T-CHO,LDL-C,HbAlc and MDA were significantly decreased(P<0.05),the SOD of rats in Rx inhibitor group were increased(P<0.05),and the MDA were decreased(P<0.05).Compared with the Rx inhibitor group,the body mass of rats in the Zicui Tongmai Yin high and low dose groups were increased(P<0.05),and the indexes of FPG,FINS,HOMA-IR,TG,T-CHO,LDL-C,HbAlc and MDA were significantly decreased(P<0.05).Compared with the Rx inhibitor group,there was no significant difference in SOD activity and MDA content between the high and low dose groups of Zicui Tongmai Yin(P>0.05).In the model group,the arrangement of myocardial fibers and myocardial cells was disordered,the fiber gap was significantly widened,the myocardial cells were hypertrophic and deformed,inflammatory cell infiltration and interstitial fibrosis were obvious.Compared with the model group,the myocardial cell arrangement,myocardial cell morphology and myocardial fiber gap in the high and low dose groups of Zicui Tongmai Yin and Rx inhibitor groups were improved.Compared with the blank control group,the relative expression of PKC-βⅡprotein in myocardial tissue of the model group was significantly higher(P<0.01).Compared with the model group,the relative expression of PKC-βⅡprotein in myocardial tissue of rats in the Zicui Tongmai Yin low-dose group and Rx inhibitor group significantly decreased(P<0.01).Compared with the model group,there was no significant difference in the relative expression of PKC-βⅡprotein in myocardial tissue of rats in the Zicui Tongmai Yin high-dose group(P>0.05).Conclusion:Zicui Tongmai Yin can improve glucose and lipid metabolism and oxidative stress in rats with diabetes cardiomyopathy,and inhibit the expression of PKC-βⅡto prevent myocardial fibrosis.
