摘要
目的:建立龙血竭药材的薄层色谱、超高效液相色谱指纹图谱和含量测定方法,为其质量标准的制定提供依据。方法:采用薄层色谱(TLC)法,以龙血竭对照药材、龙血素A及龙血素B为对照进行18批样品的定性鉴别;采用超高效液相色谱(UPLC)法同时建立龙血竭的指纹图谱和含量测定方法;使用聚类分析、主成分分析进行化学计量学的分析,建立综合评分方法,评价云南省不同产地样品的质量。结果:开发的TLC方法操作简单,能高效鉴定龙血竭药材;通过对18批样品的指纹图谱分析,识别了16个共有峰,确证了白藜芦醇、7,4’-二羟基黄酮、龙血素A、龙血素B、紫檀茋5个化学成分,并对5个成分进行定量分析,回收率在98.64%~100.89%之间;通过聚类分析,发现样品大致可分为三类,与产地因素具有一定的相关性;通过主成分分析与综合得分F结果表明整体质量最好的为第Ⅱ类。结论:建立龙血竭的薄层色谱、含量测定及指纹图谱与化学计量学相结合的方法,为龙血竭的质量评价提供新的思路,对质量标准的研究提供参考。
Objective:To establish the TLC and UPLC fingerprints and content determination methods of Longxuejie(Resina Draconis,RD),and provide the basis for the establishment of its quality standards.Methods:Qualitative identification of 18 batches of samples was carried out by TLC,taking RD as the reference material,Loureirin A and Loureirin B as the control.UPLC was used to establish the fingerprint and content determination method of RD at the same time.Cluster analysis and principal component analysis were used for chemometrics analysis,and a comprehensive scoring method was established to evaluate the quality of samples from different places in Yunnan Province.Results:The TLC was simple to operate and can identify RD efficiently.Through the fingerprint analysis of 18 batches of samples,16 common peaks were identified,and five chemical components of resveratrol,7,4'-dihydroxyflavone,loureirin A,loureirin B,and stilbene were confirmed.The five components were quantitatively analyzed,and the recovery rate was between 98.64%and 100.89%.Through cluster analysis,it was found that the samples can be roughly divided into three categories,which had a certain correlation with the factors of origin.The results of principal component analysis and comprehensive score F showed that the best overall quality was Class II.Conclusion:The establishment of the method combining with TLC,content determination,fingerprint and chemometrics of RD provides a new idea for the quality evaluation of RD and a reference for the study of quality standards.
