灵芝子实体醇提物抗缺血性脑卒中研究

Study on the Anti-Ischemic Stroke Effect of Alcohol Extract from Ganoderma lucidum Fruit Body

研究灵芝子实体醇提物对大脑中动脉闭塞/再灌注(MCAO/R)损伤模型大鼠的影响。建立SD大鼠MCAO/R大鼠模型,按行为学评分随机分为6组,每组6只,分别为灵芝子实体醇提物高剂量组(20 mg/kg)、灵芝子实体醇提物中剂量组(10 mg/kg)、灵芝子实体醇提物低剂量组(5 mg/kg)、阳性对照氯吡格雷组7.5 mg/kg和假手术组及缺血模型对照组(均给以等容积的生理盐水)。每组复灌之后2 h开始给药,之后每日灌胃给药1次,连续给药7 d。MCAO/R后第2天、第4天及末次给药后30 min,对大鼠进行神经行为学评分,测定大鼠神经功能;2,3,5-氯化三苯基四氮唑(TTC)染色法测定大鼠脑梗死率和脑含水量。用断尾法和玻片法分别检测小鼠出血时间和凝血时间。灵芝子实体醇提物在MCAO/R后2 h给药,3个剂量组MCAO/R大鼠的神经功能评分均降低脑梗死率和脑含水量,能显著延长小鼠的出血时间和凝血时间。灵芝子实体醇提物及时治疗给药可以降低大鼠脑缺血再灌注损伤脑梗死率和脑含水量,改善大鼠神经运动功能,并表现出较好的抗凝血作用。

To study the effect of alcohol extract of Ganoderma lucidum fruit body on middle cerebral artery occlusion/reperfusion(MCAO/R)injury model rats.MCAO/R model of SD rats was established and randomly divided into 6 groups according to behavioral scores,with6 rats in each group,high dose group(20 mg/kg),medium dose group(10 mg/kg)and low dose group(5 mg/kg),respectively;The positive control group was given clopidogrel 7.5 mg/kg;Sham operation group and ischemia model control group were given the same volume of normal saline.The administration of each group was started 2 h after the reperfusion,and then administered by intragastric administration once a day for 7 consecutive days.On the second day and the fourth day after MCAO/R administration and 30 min after the last administration,neurobehavioral scores were performed to determine the neural function of the rats.2,3,5-Triphenyltetrazolium chloride(TTC)staining method was used to determine the rate of cerebral infarction and brain water content in rats.The bleeding time and coagulation time of mice were detected by tail docking method and glass slide method,respectively.The alcohol extract of Ganoderma lucidum fruit body was administered 2 hours after MCAO/R,and the neurological function score,cerebral infarction rate and cerebral water content of MCAO/R rats were all decreased in the three dosage groups,and the bleeding time and coagulation time of mice were significantly prolonged.The timely treatment and administration of alcohol extract of G.lucidum fruit body can reduce cerebral infarction rate and cerebral water content,improve neuromotor function and show good anticoagulant effect in rats with cerebral ischemia-reperfusion injury.
