补骨颗粒对体内异位成骨影响研究

Effect of Bugu granules on ectopic osteogenesis in vivo

摘要 目的观察补骨颗粒对体内异位骨形成、血管形成及血管内皮生长因子(VEGF)与骨形成蛋白2(BMP-2)表达的影响,探讨补骨颗粒对骨折愈合的影响机制。方法将30只SD大鼠按照随机对照原则分为空白组、对照组、补骨颗粒组,每组10只。对照组于股四头肌肌袋中植入灌有VEGF165溶液的缓释泵,给予生理盐水灌胃;空白组和补骨颗粒组于股四头肌肌袋中植入灌有生理盐水的缓释泵,空白组给予生理盐水灌胃,补骨颗粒组给予补骨颗粒3.08 g/kg灌胃。3组均于干预2周后取肌袋区域肌肉,使用4%多聚甲醛固定并通过组织学观察成骨面积、胶原面积及CD31表达量(血管生成数);取干预前和干预2周后血清,应用ELISA法检测VEGF与BMP-2含量。结果补骨颗粒组的成骨面积明显高于空白组(P<0.05);3组间胶原面积比较差异均无统计学意义(P均>0.05);补骨颗粒组的血管生成数明显多于空白组及对照组(P均<0.05)。干预2周后,补骨颗粒组血清VEGF含量明显高于空白组(P<0.05),血清BMP-2含量明显高于对照组(P<0.05)。结论补骨颗粒可通过上调VEGF和BMP-2的表达,促进血管新生而促进体内异位成骨。 Objective It is to observe the effects of Bugu granules on bone formation and angiogenesis as well as the expression of vascular endothelial growth factor(VEGF)and bone morphogenetic protein 2(BMP-2),so as to explore the mechanism of Bugu granules on fracture healing.Methods Thirty Sprague-Dawley(SD)rats were divided into 3 groups according to the random control table,with 10 rats in each group.A slow-release pump filled with VEGF165 solution was implanted in the quadriceps muscle bag in the control group gavaged with saline gavage,a slow-release pump filled with normal saline was implanted in the quadriceps muscle bag in the blank group and Bugu granules group respectively gavaged with normal saline and Bugu granules 3.08 g/kg.In all three groups,the muscles were taken from the muscle bag area after 2 weeks of intervention,fixed with 4%paraformaldehyde,and the osteogenic area,collagen area and CD31 expression(angiogenic number)were observed by histology,the serum was taken before and after 2 weeks of intervention,and the levels of VEGF and BMP-2 l were detected by ELISA.Results The osteogenic area of the Bugu granules group was significantly higher than that of the blank group(P<0.05);the differences in collagen area among the three groups were not statistically significant(all P>0.05);the number of angiogenesis in the Bugu granules group was significantly higher than that of the blank group and the control group(both P<0.05).After 2 weeks of intervention,the level of serum VEGF in the Bugu granules group was significantly higher than that in the blank group(P<0.05),and the level of serum BMP-2 was significantly higher than that in the control group(P<0.05).Conclusion Bugu granules can promote ectopic osteogenesis in vivo by upregulating the expression of VEGF and BMP-2 and promoting angiogenesis.
