芒果食叶象甲的生物学特性及其rDNA-ITS1序列特征分析

摘要 基于前期对百色地区芒果食叶象甲种类与为害调查的基础上,对为害指数较高的芒果切叶象甲、芒果蓝卷叶象和绿鳞象甲进行了生物学特征观察,通过掌握其生活史和生活习性,以期为百色地区芒果象甲为害的监测与防治提供一定的借鉴。对13头芒果食叶象甲rDNA-ITS1基因序列进行测序与分析,结果表明:13头象甲rDNA-ITS1序列长度为988~1240 bp,其中,A+T含量(64.37%~74.89%)显著高于G+C含量(25.11%~35.63%),表现出明显的A+T偏好性特点;NJ进化树将13头象甲分为2大类群,其中,象甲1-4、1-6与其他象甲的亲缘关系较远而聚类为1个分支,象甲1-2单独为1个亚类群,其余10头象甲的亲缘关系较近而聚类为另1个亚类群。rDNA-ITS1基因序列能够真实地反映出芒果食叶象甲之间的相似性与遗传分歧,适于今后开展芒果象甲分子生物学相关研究。 Based on the previous investigation on the species and damage of the mango leaf-eating weevil in Baise area,the biological characteristics of the Deporaus marginatus Pascoe,Involvulus sp.and Hypomeces squamosus Herbst with high damage rate index were observed in this study.Through the observation of the biological characteristics of life history and living habits,in order to provide a certain reference for the monitoring and prevention of mango weevil damage in Baise area,the rDNA-ITS1gene sequences of 13 mango leaf-eating weevil were sequenced and analyzed.The results showed that the length of 13 weevil rDNA-ITS1 sequences ranged from 988 to 1240 bp,and the A+T content(64.37%~74.89%)was significantly higher than the G+C content(25.11%~35.63%),showing obvious A+T preference characteristics.The 13 weevil species are divided into 2 major groups by NJ evolutionary tree,among which weevil 1-4 and 1-6 are distantly related to other weevil species and clustered into one branch.Weevil 1-2 are a separate subgroup,and the remaining 10 weevil species are closely related to each other and clustered into another subgroup.The rDNA-ITS1 gene sequence can truly reflect the similarities and genetic differences among the mango leaf-eating weevil,which is suitable for the future research on the molecular biology of the mango leaf-eating weevil.
