多指标综合评分法结合正交试验优化黄芪-当归水提物制备工艺 被引量:2

Optimization of Preparation Technology of Aqueous Extracts of Astragalus membranaceus-Angelicae sinensis by Multi-index Comprehensive Scoring Method Combined with Orthogonal Test

摘要 为优化黄芪-当归(1∶1)水提物的制备工艺,以干浸膏得率和毛蕊异黄酮、阿魏酸及多糖占比的综合评分为评价指标,以温度、时间、料液比、提取次数为因素,在单因素试验基础上进行4因素3水平的L9(34)正交试验,优化黄芪-当归水提物制备工艺。结果显示,最优制备工艺为温度80℃、提取时间40 min、料液比1∶15、提取3次。在此条件下平均综合评分值为98.414,干浸膏得率为57.3%,阿魏酸、毛蕊异黄酮、多糖的平均占比分别为0.0579%、0.0286%和63.354%。结果表明,多指标综合评分法结合正交试验法优化黄芪当归水提物的制备工艺方法稳定可行,可为中试生产提供切实有效的理论依据。 To optimize the preparation technology of aqueous extracts of Astragalus membranaceus-Angelicae sinensis(1∶1),the yield of dry extract,the comprehensive scores of calycosin,ferulic acid and polysaccharide proportion were used as evaluation indexes,and the temperature,time,solid-liquid ratio and extraction times were used as factors.On the basis of single factor experiment,L9(3~4)orthogonal experiment with four factors and three levels was carried out to optimize the preparation process of aqueous extracts of Astragalus membranaceus and Angelicae sinensis.The results showed that the optimum preparation process was 80℃,extraction time 40 min,solid-liquid ratio 1∶15 and extraction for 3 times.Under these conditions,the average comprehensive score was 98.414,the dry extract yield was 57.3%,and the average proportions of ferulic acid,calycosin and polysaccharide were 0.0579%,0.0286%and 63.354%,respectively.It is proved that the multi-index comprehensive scoring method combined with orthogonal test is stable and feasible to optimize the preparation process of Astragalus membranaceus-Angelicae sinensis aqueous extract,which can provide practical and effective theoretical basis for pilot production.
