摘要
采用液质联用(LC-MS)多反应监测(MRM)模式,基于不同特征肽的同时测定,建立一种可以同时鉴别鹿、驴、马、牛、猪来源的胶类中药的检测方法。选择上述来源胶类中药的18条特征肽对照品,优化LC-QQQ MS的色谱、质谱条件,建立同时测定特征肽的方法,以鉴别多来源胶类中药,进一步评价检测方法的特异性、灵敏度。本方法可实现18条特征肽的同时测定,根据特征肽检测的“有”或“无”,可区分不同物种来源的胶类中药;根据特征肽相对量“高”或“低”,可区分相同物种不同部位的胶类中药,检测灵敏度可达5%。本鉴别方法特异性、灵敏度良好,为胶类中药质量控制及质量标准的提高提供了科学依据,对推动胶类中药市场健康发展具有重要意义。
Based on the simultaneous determination of various specific peptides,a method for simultaneous identifying gelatinous traditional Chinese medicines(TCMs)derived from various animal species,such as deer,donkey,cattle and pig,using liquid chromatography-mass spectrometry(LC-MS)with multi reaction monitoring(MRM)mode in the present work was established.For total 18 specific peptides selected from the animal species above,chromatographic and mass spectrometric conditions of LC QQQ MS were optimized,a method for simultaneous detecting of specific peptides was established,which could be applied to distinguish various species derived gelatinous TCMs.Furthermore,the specificity and sensitivity of the present method were evaluated.As a result,18 specific peptides could be detected simultaneously,according to the presence or absence of specific peptides,gelatin from different species could be distinguished.According to the relative contents of specific peptides in different samples,gelatins from different parts of the same species could be distinguished,and the detection sensitivity could reach 5%.This method showed good specificity and sensitivity,which provided a scientific basis for the quality control and quality standard improvement of gelatinous TCMs,and is of great significance to promote the healthy development of the market of gelatinous TCMs.
