摘要
目的建立黄精总皂苷含量测定方法。方法考察香草醛-高氯酸比色法测定黄精总皂苷的实验条件和不同提取方法对其总皂苷含量测定的影响。结果香草醛-高氯酸比色法测定黄精总皂苷含量,检测波长为550 nm,显色条件:水浴温度70℃,时间15 min,在4.03~24.18μg/mL范围内呈良好的线性关系,其回归方程为Y=0.0232X-0.0015(R^(2)=0.9985),重现性较好,RSD为1.68%,平均回收率为100.02%。结论采用水饱和正丁醇超声提取,香草醛-高氯酸比色法测定黄精总皂苷操作简便、灵敏、重现性好,可用于黄精及其炮制品的质量评价。
Objective:To establish a method for the determination of total saponins in Polygonatum.Methods:The vanillin-perchloric acid colorimetry was investigated to determine the experimental conditions for the total saponins in Polygonatum and detect the effects of different extraction methods on the determination of its total saponins.Results:The vanillin-perchloric acid colorimetry was used to determine the content of total saponins in Polygonatum.The detection wavelength was 550 nm.The coloration conditions were at the 70℃ of water bath temperature for 15 min,and the linear relationship was good in the range of 4.03 to 24.18μg/mL.The regression equation was Y=0.0232-0.0015(R^(2)=0.9985).The reproducibility was good,RSDS were 1.68%,and the average recovery rate was 100.02%.Conclusion:The vanillal-perchloric acid colorimetry is simple,sensitive and reproducible for the determination of total saponins from Polygonatum by ultrasonic extraction of water-saturation and n-butanol,and can be used for quality evaluation of Polygonatum and its products.
