基于高通量测序分析进口大麦真菌多样性

Analyzing Fungal Diversity in Imported Barley Using High-Throughput Sequencing

摘要 文章介绍了高通量测序技术在食品安全及植物病害真菌检疫中的应用。利用Illumina-Hiseq测序技术,对进口和国产大麦的真菌多样性进行了研究。从澳大利亚、法国、加拿大和国产大麦样品中共获得331054个ITS序列,系统分类学分析将其归类为6门、25纲、50目、80科、115属、158种,表明样品的真菌多样性较高。进口大麦和国产大麦的优势真菌均以子囊菌门Ascomycota和担子菌门Basidiomycota为主,两者之和占到全部真菌的97.46%~99.97%;优势菌属为李维菌属Lewia、隐球菌属Cryptococcus、Udeniomyces菌属、埃里格孢属Embellisia、锁掷酵母属Sporidiobolus、赤霉属Gibberella、蜡蚧菌属Lecanicillium、链格孢属Alternaria、红酵母属Rhodotorula、曲霉属Aspergillus、格孢腔菌属Pleospora等。韦恩图分析结果显示法国、加拿大、澳大利亚和中国的大麦真菌群落种类差异较大,每个产地的大麦真菌群落有各自的特性真菌。文章利用高通量测序建立了进口和国产大麦真菌多样性生物信息学数据库,不仅能够指导以大麦为原料的食品工业的安全生产、产品质量保障,也为进出口大麦的生物安全监测预警、风险防范及管控提供了科学的数据支持。 This study introduced the application of high-throughput sequencing techniques to food safety and the investigation of fungal disease in plant quarantine.Using Illumina Hiseq sequencing technology,a total of 331054 fungal ITS sequences were obtained from samples collected from Canada,France,Australia and China.These sequences were classified into 6 phyla,25 classes,50 orders,80 families,115 genera,or 158 species,suggesting a high fungal diversity across samples.Taxonomic analysis showed that the dominant fungals were Ascomycota and Basidiomycota,which accounted for 97.46%~99.97%of the total fungi.At the genus level,dominant fungal were Lewia,Cryptococcus,Udeniomyces,Embellisia,Sporidiobolus,Gibberella,Lecanicillium,Alternaria,Bulleromyces,Rhodotorula,and Aspergillus.Venn diagram showed that the species of barley fungal communities from France,Canada,Australia and China are quite different.Barley from each region has its own characteristic fungal species.This study examined the fungal diversity of barley imported into China,and established a bioinformatics database of barley pathogens based on high-throughput sequencing results.The results revealed the fungal community composition in imported barley,which will provide scientific data support for guiding the safe production of food industry,and for biosafety monitoring,early warning,risk prevention and control of imported barley.
