‘紫叶’酢浆草感夜性运动最适内参基因的筛选 被引量:1

Optimum Reference Gene Selection of Nyctinastic Movement in Oxalis triangularis’Purpurea’

摘要 ‘紫叶’酢浆草(Oxalis triangularis’Purpurea’)为酢浆草科(Oxalidaceae)酢浆草属(Oxalis)多年生草本植物,除具有极高观赏价值外,也是研究植物感夜性运动的理想材料。为了筛选适合于研究‘紫叶’酢浆草感夜性运动的内参基因,根据白天和夜间状态下叶枕转录组测序结果,选取6个常用内参基因ACT、EF-1α、TUA、TUB、UBC和UBQ作为候选内参基因,以‘紫叶’酢浆草的叶枕、叶柄、小叶和花4个部位白天和晚上的组织为试验材料,通过RT-qPCR对各候选内参基因的表达情况进行检测,利用3个常用内参基因分析软件geNorm、NormFinder和BestKeeper对候选内参基因的稳定性进行评估。结果表明:UBC在3个软件和稳定性综合排名中均在第二,且与各个软件的第一稳定性相近,综合稳定性最高,是研究‘紫叶’酢浆草感夜性运动最适内参基因。同时也为其他‘紫叶’酢浆草基因表达量相关研究提供参考。 Oxalis triangularis’Purpurea’is a perennial herb of the genus Oxalis belonging to the Oxalidaceae family.Being of high ornamental value,it is also a good plant material for studying nyctinastic movement.To select a suitable reference gene of Oxalis triangularis’Purpurea’for studying nyctinastic movement,six candidate reference genes,ACT,EF-1α,TUA,TUB,UBC,and UBQ,were selected from the result of transcriptome of diurnal and nocturnal pulvinus,the tissue of pulvinus,petiole,leaflet,and flower at day and night were used as experimental materials.Expression of these six candidate reference genes was detected by RT-qPCR and the stability of reference gene was analyzed by ge Norm,NormFinder,and BestKeeper.The results show that UBC ranked the second among the three software and its stability was similar to the first among all software,and its comprehensive stability was the highest.UBC was the most suitable reference gene for the study of nyctinastic movement in Oxalis triangularis’Purpurea’.It also provided reference for the other study on the gene expression of Oxalis triangularis’Purpurea’.
