茄子品种‘绿秀丽’青茄纯度鉴定的SSR标记筛选 被引量:1

摘要 本研究以茄子品种‘绿秀丽’青茄及其亲本为试验材料,用简单序列重复(simple sequence repeat,SSR)分子标记技术研究杂交种与双亲之间的扩增条带多态性以鉴定真假杂交种。结果发现,在70对SSR引物中筛选出了5对在亲本间表现差异明显,并且在杂交种中表现为双亲互补带型的引物,适合做杂交种纯度鉴定。从5对引物中选用2对引物eme01D03和ecm009对196株‘绿秀丽’青茄杂交种进行纯度分析,结果表明,eme01D03所测纯度为100.0%,ecm009鉴定的杂交种纯度为100.0%,196个单株的鉴定结果一致,一致率达到100.0%。因此,筛选出的2个SSR引物可以用于茄子杂交种‘绿秀丽’青茄种子纯度的快速鉴定。 In order to evaluate the purity of hybrid seeds,eggplant variety’Lüxiuli’green eggplant and their parental inbred lines were used to study the bands polymorphism amplified by SSR markers.The results showed that five primer pairs,namely,with obvious different bands between parents and with codominant bands in hybrid’Lüxiuli’green eggplant,were screened from 70 pairs of SSR primers,which were suitable for identification of seed purity.Then,these two SSR primers(eme01D03 and emc009)were chosen to test the purity of 196’Lüxiuli’green eggplant hybrids.The results showed that the seed purity identified by eme01D03 and ecm009 were100.0%and 100.0%,respectively,of which the genotype of 196 seedlings tested by the two SSR markers were totally identical,and the concordance of molecular test was high to 100.0%.Therefore,the screened two SSR markers could be used to quickly identify the seed purity of’Lüxiuli’green eggplant.
