枸杞多糖促进压疮大鼠皮肤创面愈合的作用机制研究 被引量:3

摘要 目的探讨枸杞多糖(LBP)对压疮(PU)大鼠创面愈合的作用机制。方法将50只Wistar大鼠随机数字表法分为5组:对照组、模型组、LBP高剂量组[100 mg/(kg·d)]、LBP中剂量组[75 mg/(kg·d)]和LBP低剂量组[50 mg/(kg·d)]。采用缺血再灌注损伤方法制备PU大鼠模型并给予LBP进行治疗2周。观察各组大鼠创面的治愈率。采集血清进行IL-6、IL-1β、TNFα和VEGF水平检测。采用RT-qPCR方法对皮肤创面组织中IL-6、TNFα和VEGF的mRNA水平进行检测。结果LBP可以增加PU大鼠创面组织愈合率,并随剂量增加愈合率增高,具有剂量依赖性。与模型组相比,LBP治疗可以明显降低血清IL-6、IL-1β和TNFα的水平,提高VEGF水平,并具有剂量依赖性,差异具有统计学意义(P<0.05)。与模型组相比,LBP治疗后PU大鼠创面组织中IL-6和TNFα的mRNA明显下降,而VEGF mRNA升高,并具有剂量依赖性,差异具有统计学意义(P<0.05)。结论LBP在PU大鼠创面愈合过程中发挥了抗炎和促进毛细血管增生的作用,为创面愈合提供了良好的内环境。 Objective To explore the mechanism of Lycium barbarum polysaccharides(LBP)on pressure ulcer(PU)wound healing in rats.Methods 50 Wistar rats were randomly divided into blank group,model group,LBP high-dose group[100 mg/(kg·d)],LBP medium-dose group[75 mg/(kg·d)]and LBP low-dose group[50 mg/(kg·d)].The PU rat model was prepared by the method of ischemia-reperfusion injury and treated with LBP for two weeks.The healing rates of the wounds of rats in each group were observed.Serum was collected for detection of IL-6,IL-1β,TNFαand VEGF levels.The mRNA levels of IL-6,TNFαand VEGF in tissues were detected by RT-qPCR method.Results LBP effectively increased the wound healing rate of PU rats in a dose-dependent manner.Compared with the model group,LBP treatment significantly reduced the levels of serum IL-6,IL-1βand TNFα,and increase the level of VEGF in a dose-dependent manner,and the differences were statistically significant(P<0.05).Compared with the model group,the mRNA of IL-6 and TNF-αin the wound tissue of PU rats after LBP treatment decreased significantly,while the mRNA of VEGF increased in a dose-dependent manner,and the differences were statistically significant(P<0.05).Conclusion LBP plays the role of anti-inflammation and promoting capillary proliferation in the process of wound healing in PU rats,and provided a good internal environment for wound healing.
