摘要
目的探讨脑出血(ICH)对干扰素调节因子4(IRF4)和炎症的影响。方法采取胶原酶Ⅳ纹状体注入Sprague Dawley(SD)大鼠右侧纹状体中建立大鼠ICH模型,通过蛋白印迹(Westernblotting)和ELISA等方法测定ICH后48h大鼠的IRF4的表达及炎症变化。利用血红素(Hemin)诱导大鼠嗜铬细胞瘤细胞(PC12)损伤模拟体外ICH模型,并检测IRF4及炎症变化;在PC12细胞中,采用短发夹RNA(shRNA)-IRF4沉默IRF4后,检测其对炎症反应的影响。结果手术后48h ICH行为学评分最低。ICH组IRF4,IL-6,IL-1β表达水平显著升高。结论ICH可以促进IRF4的表达和炎症反应,降低IRF4的表达可减缓炎症反应。
Objective To investigate the effect of intracerebral hemorrhage(ICH)on interferon regulatory factor 4(IRF4)and inflammation.Methods the ICH model was established by injecting collagenaseⅣstriatum into the right striatum of SpragueDawley rats.The expression and inflammation of IRF4 in 48 hours after ICH were measured by Western-blotting and ELISA.The injury of rat pheochromocytoma cell cells was induced by hemin to simulate ICH model in vitro,and IRF4 and inflammatory changes were detected;short hairpin RNA(shRNA)-IRF4 was used to silence the expression of IRF4 in PC12 cells and its effect on inflammation.Results The behavioral score of cerebral hemorrhage was the lowest 48 hours after operation.The expression of IRF4 in ICH group was significantly higher than that in sham operation group.IL-6,IL-1 in ICH groupβThe expression was significantly increased.Conclusion ICH can promote the expression and inflammatory response of IRF4,and reduce the expression of IRF4 can slow down the inflammatory response.
