甲状腺细针穿刺不同细胞学标本保存方法对BRAF V600E基因突变检测的影响 被引量:1

摘要 目的:探讨不同保存方法对甲状腺细针穿刺细胞学标本BRAF V600E基因突变检测的影响。方法:回顾性分析自2019年01月至2022年02月华北理工大学附属医院收治的接受B超引导下行甲状腺细针穿刺检查的患者164例。分别从商品化细胞保存液固定标本、生理盐水保存标本和刮取染色后涂片标本中提取DNA并检测BRAF V600E基因突变。结果:通过三种保存方法提取的DNA浓度均数及内控Ct值相比,差异均无统计学意义(P>0.05)。细胞保存液组和生理盐水保存液组、生理盐水保存液组和刮片组所提取DNA的A260/280值相比,差异均无统计学意义(P>0.05),而细胞保存液组和刮片组所提取DNA的A260/280值比较,差异有统计学意义(P<0.05)。结论:甲状腺FNAC涂片标本刮片后进行基因突变检测获得DNA的纯度高,且对实验室要求低,适宜在基层医院推广。 Objective:To investigate the effect of different methods of preservation of detection of BRAF V600E mutation in fine needle aspiration cytology(FNAC)of thyroid.Methods:Retrospective analyzed study of 164 patients from Jan 2019 to Feb 2022 who underwent thyroid fine needle aspiration through ultrasound-guided in our hospital.DNA extracted respectively from materials preserved in commercial cell preservation solution,normal saline preservation solution and stained smears is used for the detection of BRAF V600E mutation.Results:There are no significant differences in the mean DNA concentration and the Ct value of reference gene among the three methods(P>0.05).For the A260/280value,there was no statistically significant difference between the cell preservation solution group and the saline preservation solution group,nor between the saline preservation solution group and the scraping slice group(P>0.05),while there was statistically significant difference in the A260/280value between the cell preservation solution group and the scraping slice group(P<0.05).Conclusion:Scraping thyroid FNAC samples can obtain high DNA purity and low laboratory requirements,which is suitable for promotion in primary hospitals.
