息半夏种质资源遗传多样性的ISSR分析

ISSR Analysis of Genetic Diversity of Xi Pinellia Ternata Germplasm Resources

对信阳息县道地药材息半夏种质资源遗传多样性进行研究,利用ISSR技术对24份半夏进行扩增。共挑选出了8条扩增条带清晰的引物,并检测到117条明显的条带,多态性条带为110条,平均多态性信息指数(PIC)为0.8508,平均观测等位基因数(Na)为1.9279,平均有效等位基因数(Ne)为1.4811,平均Nei氏基因多样性(H)为0.2927,平均香农信息指数(I)为0.4481。利用UPGMA方法进行聚类分析,24份半夏在遗传相似系数0.5901处被分为3个类群。结果表明,24份半夏种质资源具有较高的遗传多样性。

In order to study the genetic diversity of genuine medicinal materials Xi Pinellia ternata(Thunb.)Breit.In Xinyang Xi County,ISSR is used to amplify the 24 P.ternata.8 ISSR primers with clear amplification bands are selected,117 obvious bands are detected,of which 110 bands are polymorphic,and the mean polymorphism information content(PIC)is 0.8508.The average observed number of alleles(Na),effective number of alleles(Ne),Nei’s genetic diversity index(H)and Shannon’s information index(I)are 1.9279,1.4811,0.2927 and 0.4481,respectively.Cluster analysis by UPGMA shows that 24 P.ternata are divided into three groups at a genetic similarity of 0.5901.The results show that there are high genetic diversity among 24 P.ternata.
