硫氧还蛋白5在精索静脉曲张大鼠睾丸中的表达及意义

摘要 目的:观察精索静脉曲张(Varicocele, VC)大鼠以及通过高位结扎精索静脉术后大鼠睾丸组织中硫氧还蛋白5 (TXNDC5)的表达水平。方法:将30只7周龄雄性SD大鼠按照随机数字表法平均分入模型组、假手术组和治疗组。模型组大鼠按照经典的Turner法建立精索静脉曲张模型;假手术组大鼠仅游离左精索静脉而不进行缩窄和结扎;治疗组大鼠在模型组造模的基础上在术后8周行左精索静脉高位结扎术。三组大鼠继续饲养4周后留取标本。对各组大鼠睾丸组织进行HE染色,观察睾丸组织病理形态;免疫组化检测TXNDC5的蛋白表达情况。结果:假手术组睾丸组织形态正常,而模型组和治疗组大鼠睾丸组织均存在不同程度的损伤,且治疗组损伤程度较低。治疗组大鼠睾丸组织内TXNDC5的蛋白表达量比模型组升高但是仍低于假手术组。结论:精索静脉曲张可以导致睾丸组织损伤,使睾丸组织TXNDC5表达量下降,而精索静脉曲张高位结扎可以一定程度扭转精索静脉曲张引起的损伤。 Objective: To observe the expression level of Thioredoxin domain containingprotein 5(TXNDC5) in the testis of Varicocele(VC) rats and rats treated with high spermatic vein ligation. Methods:Thirty 7-week-old male Sprague-Dawley rats were randomly divided into model group, sham operation group and treatment group. In model group, varicocele model was established according to classic Turner method. In sham operation group, the left spermatic vein was isolated without constriction and ligation. On the basis of the model group, the rats in the treatment group were subjected to high ligation of the left spermatic vein 8 weeks after operation.The rats in the three groups were kept for 4 weeks and the specimens were collected. The testicular tissue of rats in each group was stained with HE, and the pathological conditions of each testicular tissue were observed. The protein expression of TXNDC5 was detected by immunohistochemistry. Results:The testicular tissue of the sham operation group was normal, while the testicular tissue of the model group and the treatment group had different degrees of damage, and the treatment group had a lower degree of damage. The protein expression of TXNDC5 in testicular tissue of rats in treatment group was higher than that in model group, but still lower than that in sham group. Conclusion:Varicocele can cause testicular tissue injury and decrease the expression of TXNDC5 in testicular tissue. High ligation of varicocele can reverse the injury caused by varicocele to some extent.
