黏附性聚多巴胺@己脒定二羟乙基磺酸盐微胶囊的制备及其性能研究

摘要 为提高去屑剂己脒定二羟乙基磺酸盐(HD)在湿润环境下的黏附性能,以W_(1)/O/W_(2)双重乳液为模板,合成具有黏附性的聚多巴胺@己脒定二羟乙基磺酸盐微胶囊(PDA@HD微胶囊),并对其形貌结构、载药性能、缓释性能、黏附性能及斑马鱼胚胎急性毒性进行研究。结果表明:PDA@HD微胶囊外观呈球状,有明显核壳结构,平均粒径为353 nm;通过提高双重乳液W_(1)相中HD的质量浓度,可以显著提升PDA@HD微胶囊中HD的搭载率,当HD质量浓度为18 mg/mL,其搭载率为17.1%;HD在PDA@HD微胶囊中的释放表现出明显的pH依赖性,偏酸性环境更有利于HD的释放;PDA@HD微胶囊的黏附力提高明显,其平均黏附力(8.42 nN)约为HD(2.89 nN)的3倍;PDA@HD微胶囊的斑马鱼胚胎急性毒性有所降低,当PDA@HD微胶囊内的HD质量浓度为0.5 mg/mL时,斑马鱼胚胎发育至96 hpf仍无死亡。 Adhesive polydopamine nanocapsules containing hexamidine diisethionate(PDA@HD nanocapsules)were prepared using W_(1)/O/W_(2)emulsion templating to enhance wet adhesion capacity of HD.Thereafter,the morphological structure,loading and release properties,adhesion properties and acute toxicity to zebrafish embryos of PDA were investigated.The results showed that the PDA@HD nanocapsules were spherical with a distinct core-shell structure whose average particle size was 353 nm.The loading ratio of PDA@HD nanocapsules could be effectively increased by increasing the concentration of HD in W_(1)phase of W_(1)/O/W_(2)emulsion.Specifically,by increasing the concentration of HD to 18 mg/mL,the loading ratio of PDA@HD nanocapsules increased to 17.1%.The release of HD in PDA@HD nanocapsules exhibited a clear pH dependence and PDA@HD nanocapsules showed better release properties in acidic environment.The adhesion force of PDA@HD nanocapsules was effectively increased about 3 times that of HD.The PDA@HD nanocapsules reduced the acute toxicity to zebrafish embryos.When the concentration of HD in PDA@HD nanocapsules was 0.5 mg/mL,all zebrafish embryos survived till 96 hpf.
