辣木叶多酚的超声提取及抗氧化活性分析 被引量:2

摘要 以乙醇为提取溶剂,超声辅助提取法提取辣木叶中多酚类物质。采用单因素试验结合BoxBenhnken响应曲面设计优化提取条件。根据最佳工艺参数,测定不同产地辣木叶多酚的含量及抗氧化活性,采用ABTS、FRAP方法测定辣木叶多酚的化学抗氧化活性,采用MTT法测定辣木多酚对过氧化氢诱导的PC-12细胞氧化损伤的保护作用。实验结果表明,超声辅助提取法最佳提取工艺为:乙醇浓度80%,料液比1∶30(g/mL),温度51℃,时间52 min;不同产地总多酚含量:欧洲辣木>印度辣木>非洲辣木>云南辣木,欧洲辣木叶的多酚总含量最高,为29.11 mg GAE/g DW。四个产地的体外抗氧化实验结果表明,欧洲辣木叶多酚表现出较强的抗氧化活性。本实验建立了一种辣木叶多酚提取方法,辣木叶多酚是一种潜在的天然抗氧化剂,具有很高的应用价值及开发前景。 Ultrasonic-assisted extraction was used to extract polyphenols from Moringa oleifera leaves with ethanol as solvent. Single-factor experiment combined with Box-Benhnken response surface method were used to optimize the extraction conditions. According to the optimum extraction conditions,the content of polyphenol extracted from Moringa oleifera leaves of different geographic origins were determined. The antioxidant activity of polyphenol from different Moringa oleifera leaves was determined by ABTS and FRAP methods in vitro,and the cellular antioxidant activity was measured on PC-12 cells by MTT method. The experimental results showed that the optimal ultrasonicassisted extraction conditions were ethanol concentration 80%,solid-liquid ratio 1∶30(g/mL),temperature 51℃,time 52 min. Total polyphenol content extracted from Moringa oleifera leaves of different geographic origins :Europe Moringa oleifera > India Moringa oleifera > Africa Moringa oleifera > Yunnan Moringa oleifera,and total polyphenol content extracted from Europe Moringa oleifera leaves was the highest,reached 29.11 mg GAE/gDW. The antioxidant activity analysis in vitro of polyphenol extracted from Moringa oleifera leaves of four geographic origins showed that polyphenol from leaves of Europe Moringa oleifera showed strong antioxidant activity. In this study,an extraction method of Moringa oleifera leaves polyphenols was established and it was found that Moringa oleifera leaves polyphenols could be used as a natural antioxidant with high application value and development prospect.
