诱导自噬对朊病毒病治疗、作用机制的研究进展

摘要 自噬作为人体基本生理、病理过程,在朊病毒病的发生、发展以及治疗中都不容忽视,但其所起到的作用和机制至今尚未完全阐明。近几年来,许多研究都着力于通过增强自噬,加强对异常朊蛋白的清除,抑制相应毒性朊蛋白的聚集,降低正常朊蛋白的转化率来延缓疾病进程,最终达到治疗朊病毒病的目的。文章针对自噬在朊病毒病中的作用机制、朊病毒病相关的药理学与病理学机制以及相关药物治疗进行总结。并对通过诱导自噬靶向治疗朊病毒病的可行性进行讨论和评价,探索诱导自噬作为治疗朊病毒病的有效途径,旨在为深入认识自噬在朊病毒病治疗领域中的意义提供参考。 As a basic physiological and pathological process in human body,autophagy cannot be ignored in the occurrence,development and treatment of prion diseases,but its role and mechanism have not been fully elucidated yet.In recent years,many studies have focused on the enhancement of autophagy,strengthening the clearance of abnormal prions,inhibiting the aggregation of corresponding toxic prions,reducing the conversion rate of normal prions,to slow down the disease process and ultimately achieve the goal of treating prion diseases.The article summarizes the mechanism of autophagy in prion diseases,the pharmacological and pathological mechanisms related to prion diseases,and the related drug therapy.It also discusses and evaluates the feasibility of targeting the treatment of prion diseases through induced autophagy,and explores induced autophagy as an effective way to treat prion diseases,aiming to provide a reference for an in-depth understanding of the significance of autophagy in the field of prion disease treatment.
